Low-power laser irradiation promotes the proliferation and osteogenic differentiation of human periodontal ligament cells via cyclic adenosine monophosphate

Abstract: Retaining or improving periodontal ligament (PDL) function is crucial for restoring periodontal defects. The aim of this study was to evaluate the physiological effects of low-power laser irradiation (LPLI) on the proliferation and osteogenic differentiation of human PDL (hPDL) cells. Cultured hPDL cells were irradiated (660 nm) daily with doses of 0, 1, 2 or 4 J⋅cm−2. Cell proliferation was evaluated by the 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) assay, and the effect of LPLI on ost… Show more

“…Reverse transcription PCR was performed using Primescript RT reagent Kit (TaKaRa), and further quantitative real-time PCR analysis was performed using SYBR select master mix (Applied Biosystems; TCAB1 primers: Forward, 5’-CATATCTGGGACGCATTCACT-3’, Reverse, 5’-GTTGAAGCCACAGAAGAGCTG-3’). The PCR results were quantified using semi-quantitative method as previously reported [22]. …”

“…The second route involves the well-known ability of PBM to increase the expression levels of cAMP [9092]. As mentioned above, cAMP activates protein kinase A which then phopsphorylates AANAT thus preserving its enzyme bioactivity and increasing melatonin synthesis.…”
