Lower Proportion of CD45R0+ Cells and Deficient Interleukin-10 Production by Formula-Fed Infants, Compared With Human-Fed, Is Corrected With Supplementation of Long-Chain Polyunsaturated Fatty Acids

Abstract: Adding LCP to a preterm infant formula resulted in lymphocyte populations, phospholipid composition, cytokine production, and antigen maturity that are more consistent with that in human milk-fed infants. This may affect the ability of the infant to respond to immune challenges.

“…Isolated cells (200 000 cells per well) were used to determine the cell phenotype using a two-colour direct immunofluorescence (Field et al, 2000). Antibodies used were CD3, CD4, CD8, CD25, CD28, CD62L, CD71, CD80, CD45RA (BD Bioscience, Mississauga, ON, Canada) and OX12 (Cedarlane, Hornby, ON, Canada); Streptavidin QR (Sigma, Oakville, ON, Canada) was added to all biotin-labelled antibodies.…”

The percentage of cells expressing each antibody marker was determined by flow cytometry (FacScan, Becton Dickinson, Sunnyvale, CA, USA) (Field et al, 2000). It was not always possible to perform every phenotype assay on each rat because of the variation in total yield of tissue; n for each assay is indicated in result tables.…”

“…25 The following pre-labelled monoclonal antibodies were used: CD3 and CD28 (FITC-labelled); CD4, CD8, CD3, CD11b/c and OX12 (PE-labelled); and CD25 and CD8 (biotin-labelled). All antibodies were purchased from BD Biosciences, PharMingen (Mississauga, ON, Canada) except CD4, and OX12, which were purchased from Serotec (Cedarlane Laboratories Ltd, Hornby, ON, Canada).…”

“…Binding of MKPs to their MAPK substrates increases phosphatase activity (Camps et al, 1998a;Hutter et al, 2000;Slack et al, 2001) and this is due to stabilization of the active enzyme-substrate complex (Field et al, 2000). MKP-3 exhibits high fidelity to its substrate ERK1/2 and upon binding its catalytic activity is enhanced (Camps et al, 1998b).…”
