LRH-1 mediates anti-inflammatory and antifungal phenotype of IL-13-activated macrophages through the PPARγ ligand synthesis

Abstract: Liver receptor homologue-1 (LRH-1) is a nuclear receptor involved in the repression of inflammatory processes in the hepatointestinal tract. Here we report that LRH-1 is expressed in macrophages and induced by the Th2 cytokine IL-13 via a mechanism involving STAT6. We show that loss-of-function of LRH-1 in macrophages impedes IL-13-induced macrophage polarization due to impaired generation of 15-HETE PPARγ ligands. The incapacity to generate 15-HETE metabolites is at least partially caused by the compromised r… Show more

“…Of interest, LRH-1 expression was not completely absent, suggesting that the remaining LRH-1 expression is derived from a different cellular source, e.g. as previously shown by us for T cells [32] and others for macrophages [33]. While the inଏammation-driven induction of the steroidogenic enzymes Cyp11b1, Cyp11a1 and Cyp21 was evident in control animals, epithelial-speciଏc deletion of LRH-1 resulted in a signiଏcant reduction of the expression of these enzymes ( Fig.…”

“…4E). PPAR␥ is a key transcription factor necessary for M2 differentiation, and its expression is induced by IL4, likely in a STAT6 dependent manner (17)(18)(19). We hypothesized that promoter histone arginine methylation can regulate PPAR␥ expres- sion in IL4-differentiated cells.…”

“…Finally, cytochrome P450 enzymes (e.g. CYP1A1 and CYP1B1) represent another pathway that metabolizes AA or AA-derived mediators, thus further enhancing the immunoregulatory repertoire of eicosanoids (Nebert and Karp, 2008;Divanovic et al, 2013;Lefèvre et al, 2015). Indeed, there is a large body of literature describing the regulation and immunological functions of eicosanoids and not all studies could be covered in this review.…”
