MMP13 Contributes to Dental Caries Associated with Developmental Defects of Enamel

Abstract: The aim of this study was to investigate the association between genetic polymorphisms in MMP8, MMP13, and MMP20 with caries experience and developmental defects of enamel (DDE) in children from the Amazon region of Brazil. Den tal caries and DDE data were collected through clinical examination from 216 children. Genomic DNA was extracted from saliva, and genotyping of selected polymorphisms in MMP8 (rs17099443 and rs3765620), MMP13 (rs478927 and rs2252070), and MMP20 (rs1784418) was performed using TaqMan che… Show more

“…This resulted in 21 publications including a total of 24 studies that met the eligibility criteria and were included in our analyses 5, 10-12, 21-38 . In summary, the meta-analyses of rs12640848 in ENAM included seven studies with a total of 1,256 subjects in the case group and 710 subjects in the control group 12, 25, 27, 33, 34, 37 ; the meta-analyses of rs1784418 in MMP20 included five studies with a total of 699 subjects in the case group and 817 subjects in the control group 10, 24, 26, 30, 36 ; the meta-analyses of rs17878486 in AMELX included four studies with a total of 249 subjects in the case group and 193 subjects in the control group 10, 12, 22, 25 ; and the meta-analyses of rs3796704 in ENAM included four studies with a total of 574 subjects in the case group and 533 subjects in the control group 12, 25, 32, 34 . Data for other genetic variants came from fewer studies.…”

“…The second subset included pre-adolescents aged 9 to 12 years recruited from four public schools in Manaus, state capital Amazonas, Brazil. 26,27 All children who met the inclusion criteria and whose the parents/legal guardians agreed to participate, were included. During the clinical examination, those preschool children with dental treatment needs were referred to dental treatment.…”

“…The matrix metalloproteinases (MMPs) constitute a family of secreted and membrane-associated zinc-dependent endopeptidases that are the major regulators of extracellular matrix turnover and are capable of selectively degrading a wide spectrum of both extracellular matrix and non-matrix proteins [20,21]. Matrix metalloproteinase 2, also known as gelatinase A, is a membrane-bound protein with collagen-degrading ability [20,22] and has important roles in tooth formation and mineralization [23][24][25][26][27].…”

“…Matrix metalloproteinase 2, also known as gelatinase A, is a membrane-bound protein with collagen-degrading ability [20,22] and has important roles in tooth formation and mineralization [23][24][25][26][27]. Genetic polymorphisms in enamel formation genes could contribute to structural alterations that could lead to enamel porosity, presence of enamel crystal inhibitory proteins and decreased mineral content [21,28,29]. Some studies have demonstrated the expression of MMP2 during enamel formation [23][24][25].…”
