S-genotype identification based on allele-specific PCR in Japanese pear

Abstract: Gametophytic self-incompatibility in Japanese pear (Pyrus pyrifolia Nakai) is controlled by the single, multi-allelic S-locus. Information about the S-genotypes is important for breeding and the selection of pollen donors for fruit production. Rapid and reliable S-genotype identification system is necessary for efficient breeding of new cultivars in Japanese pear. We designed S allele-specific PCR primer pairs for ten previously reported S-RNase alleles (S1–S9 and Sk) as simple and reliable method. Specific nu… Show more

“…1 ). Several methods using DNA markers have been developed that are capable of specifically discriminating S 1 – S 9 , S k , and S 4 sm ( Ishimizu et al 1999 , Kim et al 2007 , Nashima et al 2014 , 2015 , Okada et al 2008 , Takasaki et al 2004 ).…”

“…Identification of the S-genotype is important for commercial fruit production and cross breeding. Several identification systems for rapid and reliable S-genotyping have been established, such as PCR-restriction fragment length polymorphism (PCR-RFLP) analysis (Ishimizu et al, 1999) and allele-specific PCR amplification (Nashima et al, 2015). A self-compatible mutant 'Osa-Nijisseiki' derived from a self-incompatible cultivar 'Nijisseiki' possesses the S 4 sm haplotype controlling selfcompatibility.…”

“…Allele-specific primers were applied for identifying the single alleles (S1, S6, S7, S8). Nucleotide sequences of PyrusS1, PyrusS6, PyrusS7, and PyrusS8 primers were obtained using Ishimizu et al (1999) and Nashima et al (2015), respectively (Table 1). Optimization of PCR (BioRad T100 TM ) was conducted for used primers as follows: 5X PCR reaction buffer, 25 mM MgCl2, 100 µM dNTP, 5 pmol primer, 5U Taq polymerase and 50-250 ng of genomic DNA was performed at a total volume of 15 µL.…”
