Background The effect of circular RNAs (circRNAs) is widely studied in various human cancers, including breast cancer (BC). Herein, circUSPL1 has been recognized as a new regulator for BC progression. However, the detailed biological function and molecular mechanism of circUSPL1 in BC remain vague. Methods The expression level of circUSPL1, miR‐1296‐5p and metastasis associated 1 (MTA1) was examined by quantitative reverse transcription PCR. BC cell proliferation, migration, invasion, apoptosis and aerobic glycolysis were analyzed by colony formation assay, 5‐ethynyl‐2′‐deoxyuridine assay, wound healing assay, transwell assay, flow cytometry and glycolysis corresponding kits, respectively. The protein level of Bcl‐2, Bax, HK2, GLUT1 and MTA1 was evaluated by western blot analysis. The relationship of miR‐1296‐5p and circUSPL1 or MTA1 was affirmed using dual‐luciferase reporter or RIP assays. A murine xenograft model was conducted to analyze the tumor growth in vivo. Results CircUSPL1 and MTA1 expression level was increased, but miR‐1296‐5p was particularly reduced in BC tissues and cells. CircUSPL1 deficiency significantly inhibited BC cell proliferation, migration, invasion, glycolysis, and promoted cell apoptosis. In addition, circUSPL1 directly targeted miR‐1296‐5p, and downregulation of miR‐1296‐5p eliminated the inhibitory action of circUSPL1 knockdown. Additionally, overexpression of miR‐1296‐5p repressed cell malignant properties, while the suppressive effects were overturned by MTA1 elevation. Lastly, silencing of circUSPL1 inhibited tumor growth by sponging miR‐1296‐5p and regulating MTA1. Conclusion CircUSPL1 deficiency repressed BC cell malignant phenotypes through reducing MTA1 via targeting miR‐1296‐5p, which might provide a theoretical basis for BC treatment.

Role and mechanism of Circ‐PDE7B in the formation of keloid

The excessive proliferation of keloid fibroblasts is one of the important reasons leading to the formation of keloids. Circular RNA (circRNA) is an important regulator that regulates the biological functions of cells. However, the role and mechanism of circ‐PDE7B in keloid formation have not been studied yet. QRT‐PCR was used to detect the circ‐PDE7B, miR‐331‐3p and cyclin‐dependent kinase 6 (CDK6) expression. The biological functions of keloid fibroblasts were determined by MTT assay, flow cytometry, transwell assay and wound healing assay. Western blot analysis was used to measure the protein levels of extracellular matrix (ECM) markers and CDK6. The interaction between miR‐331‐3p and circ‐PDE7B or CDK6 was confirmed by dual‐luciferase reporter assay and RIP assay. Circ‐PDE7B was found to be upregulated in keloid tissues and fibroblasts. Downregulation of circ‐PDE7B could suppress the proliferation, invasion, migration, ECM accumulation and accelerate the apoptosis of keloid fibroblasts. Circ‐PDE7B could serve as a sponge of miR‐331‐3p, and the regulation of silenced circ‐PDE7B on the biological functions of keloid fibroblasts could be abolished by miR‐331‐3p inhibitor. Additionally, CDK6 was a target of miR‐331‐3p, and its overexpression could reverse the negative regulation of miR‐331‐3p on the biological functions of keloid fibroblasts. Circ‐PDE7B sponged miR‐331‐3p to positively regulate CDK6 expression. Taken together, circ‐PDE7B promoted the proliferation, invasion, migration and ECM accumulation of keloid fibroblasts by regulating the miR‐331‐3p/CDK6 axis, suggesting that circ‐PDE7B might be a potential target for keloid treatment.

BackgroundCircular RNAs (circRNAs) are closely associated with the development of breast cancer (BC). In this study, we aimed to clarify how differentially expressed circRNAs affect the development of BC.MethodsQuantitative real‐time polymerase chain reaction (qRT‐PCR) was used to detect the expression of circADAM9, miR‐1236‐3p and fibroblast growth factor 7 (FGF7). Colony formation, 5‐ethynyl‐2′‐deoxyuridine (EdU), wound healing, transwell, and flow cytometry were used to assess cell proliferation, migration, invasion, and apoptosis. Glucose consumption, lactic acid production and ATP levels were assessed using glycolysis metabolism analysis. Dual‐luciferase reporter assay and RNA immunoprecipitation (RIP) assay were carried out to verify the relationship between miR‐1236‐3p and circADAM9 or FGF7. The roles of cirADAM9 on tumor growth were analyzed using a xenograft tumor model. Ki‐67 and FGF7 expression was measured via immunohistochemistry (IHC) assay. Apoptosis‐related proteins and exosome markers were detected by western blot.ResultsCircADAM9 was highly expressed in BC cells, and circADAM9 silencing inhibited BC cell proliferation, migration, invasion, and glycolysis, and promoted cell apoptosis. Furthermore, miR‐1236‐3p inhibition could overturn circADAM9 knockdown‐mediated BC inhibition. Moreover, the negative influences of miR‐1236‐3p overexpression on BC progression were restrained via FGF7 overexpression. CircADAM9 silence also inhibited BC tumor growth in vivo.ConclusionCircADAM9 promoted BC development partly by the miR‐1236‐3p/FGF7 axis, highlighting a potential prognostic biomarker and therapeutic target for BC patients.
