Fasting Serum Levels of Potassium and Sodium in Relation to Long-Term Risk of Cancer in Healthy Men

Abstract: Purpose: To examine whether serum levels of potassium and sodium were associated with long-term cancer risk in initially healthy men. Patients and Methods: A cohort of 1994 initially healthy men with no use of medication, aged 40-59 years, was followed for cancer during 40 years of follow-up. Associations between fasting electrolyte levels and cancer risk were assessed with incidence rates and Cox proportional hazards models. Results: Potassium, but not sodium, was linearly associated with cancer risk. This as… Show more

“…(2) There may be some inherent individual factors that help regulate serum potassium, which also helps tumor progression. For example, genetic variations in ion channels and ion pumps involved in potassium homeostasis are associated with genes that regulate cell proliferation and differentiation 39 , 40 . Serum phosphate is an essential nutrient for the synthesis of nucleic acids, phospholipids, and high-energy metabolites such as adenosine triphosphate (ATP); thus, rapidly dividing cells require a continuous supply of phosphate 41 , 42 .…”

“…Notably, TIMP-1 is located on the X-chromosome and has been linked to sexspecific/restricted cancers in humans 32 . We further show that exposed females have increased levels of potassium in their blood, which also has been associated as a long-term risk factor for cancer 33 . The altered blood potassium levels might be a consequence of altered sex hormone balance as both progestin and testosterone possess potassium sparing effects 34 and this could be a consequence of the disturbed expression of genes regulating salt and water homeostasis (Ace, Ace3, Nppa, Guca2b, Shank2, Nherf2, Hsd11b2) noticed here in female rats.…”

“…For every 1 SD increase in potassium, the risk of overall cancer increased by 16%. Higher levels of serum potassium may contribute to cancer development and growth through immune mechanisms ( 121 ). Kv channel blocker 4-AP can inhibit the secretion of IL-6 and IL-1, thereby inhibiting glioma cell proliferation ( 110 ).…”
