Role of Melatonin in the Regulation of Pain

Abstract: Melatonin is a pleiotropic hormone synthesized and secreted mainly by the pineal gland in vertebrates. Melatonin is an endogenous regulator of circadian and seasonal rhythms. Melatonin is involved in many physiological and pathophysiological processes demonstrating antioxidant, antineoplastic, anti-inflammatory, and immunomodulatory properties. Accumulating evidence has revealed that melatonin plays an important role in pain modulation through multiple mechanisms. In this review, we examine recent evidence for… Show more

“…In the present study, patients receiving melatonin needed a lower dose of analgesia, and this decrease in analgesic dose increased with increasing melatonin dose. The finding of our study seems more logical considering that the analgesic effect of melatonin is dose-dependent and increases with increasing dose [ [35] , [36] , [37] ].…”

“…During day, the exposure to light induces the breakdown of melanopsin in retinal photoreceptive ganglion cells, which signals through retinohypothalamic pathway to inhibit the synthesis of melatonin, consequently, the levels of melatonin are low during daylight and rise in dark at night [3]. The neurotransmitters such as noradrenaline and GABA regulate the synthesis of Melatonin [29]. The presence of 5-methoxy and N-acetyl functional groups in the structure of melatonin impart specificity and amphiphilicity, and melatonin can readily cross the plasma membranes affecting almost all the cells of the body [3].…”

“…The pain-relieving effects of melatonin either stem from its direct anti-nociceptive effects due to its anti-oxidant, anti-inflammatory properties and direct interaction with its cognate receptors or through indirect complex interactions with other systems that can modulate the pain perception pathways [29,37].…”

“…Melatonin is a neurohormone produced by the pineal gland. This molecule has many beneficial activities, including immunomodulatory, anti-inflammatory, antioxidant properties within the body [ 158 , 159 ]. Because of these functions, some researchers have proposed this agent could be a therapeutic option for treating viral infections and respiratory diseases, including ARDS and acute lung injury (ALI) [ 160 ].…”
