Lysis of human fibroblast colony-forming cells and endothelial cells by monoclonal antibody (6-19) and complement

Abstract: The murine IgG2a monoclonal antibody 6–19 binds to a wide variety of nonhematopoietic cells including human marrow-derived stromal cells but does not bind to marrow or peripheral blood cells. We studied the effects of this antibody and rabbit complement on marrow cells. Fibroblast colonies were eliminated from light density marrow cells by a single incubation with monoclonal antibody 6–19 and complement. The growth and composition of granulocytic and erythroid colonies were unaffected. Specific complement medi… Show more

“…For example, the monoclonal antibodies 6-19 and 1B10 fixed complement have been used to remove fibroblasts from epithelial cell culture. 22 , 23 This study used antibody 1B10 to reduce or eliminate fibroblasts in human skeletal muscle primary culture. 1B10 is specific to fibroblasts and smooth muscle differentiated fibroblasts within the context of vascular smooth muscle cells.…”

“…Various approaches have been recruited to eliminate fibroblasts from cell culture, of which, complement-mediated cytotoxicity using an anti-fibroblast antibody [ 35 , 36 ] and pre-plating have been frequently used [ 32 , 37 ]. Herein, we used pre-plating on a gelatin-coated surface to decrease fibroblasts.…”
