Lysophosphatidic acid activates telomerase in ovarian cancer cells through hypoxia‐inducible factor‐1α and the PI3K pathway

Abstract: Telomerase is reactivated in over 90% of tumors and plays critical roles in tumor progression. The mechanisms by which telomerase is up-regulated in cancer cells are poorly understood. Here we showed that a bioactive lipid, lysophophatidic acid (LPA), up-regulated the expression of human telomerase reverse transcriptase (hTERT) and telomerase activity in serous ovarian adenocarcinoma cell lines SKOV3, A2780, and HEY, but not in OCC1, a clear cell ovarian cancer cell line. This cell type specific effect of LPA … Show more

“…[25,26] Furthermore, HIF-1α resulted in elongation of telomeres as a key transactivator for the induction of TERT transcription. [27] Simultaneously, some SNPs in ACYP2 have been identified to be associated with telomere length. [7,9,28,29] Subsequently, we performed a multivariate analysis adjusted for age and gender among HAPE patients and controls to investigate the association between ACYP2 genetic polymorphism and HAPE risk.…”

“…Exposure to hypoxia has been shown to activate telomerase due to its ability to increase TERT expression, which in turn results in telomere elongation [17,18]. The mechanisms responsible for hypoxia-induced TERT expression have been investigated, and it was shown that TERT expression was upregulated following the induction of HIF-1α during exposure to hypoxia and HIF-1α acted as an essential transactivator to induce TERT transcription [19-21]. …”

“…As well as inhibition of apoptosis, one of the hallmarks of cancer is the avoidance of senescence. LPA suppresses p53-dependant replicative senescence (Kortlever et al 2008), at least in part through induction of telomerase (Bermudez et al 2007;Yang et al 2008). Along with many other chemotherapeutic agents, carboplatin and paclitaxel induce apoptosis.…”
