Lysosomal Dysfunction and Autophagy Blockade Contribute to MDMA-Induced Neurotoxicity in SH-SY5Y Neuroblastoma Cells

Abstract: Methylenedioxymethamphetamine (MDMA) is a psychostimulant with high abuse potential and severe neurotoxicity. According to our previous study, MDMA promotes autophagosome accumulation and contributes to cell death in cultured cortical and serotonergic neurons. However, the detailed mechanism underlying autophagy dysfunction remains unclear. Lysosomes play an important role in autophagic degradation. The present study aimed to examine the role of lysosomal function in autophagic flux in neuronal cultures expose… Show more

“…3,4-methylenedioxymethamphetamine (MDMA) is a drug of abuse that can have more severe effects in certain environmental conditions, including elevated temperatures 51 . SH-SY5Y cells have been used previously to study the effects of MDMA, so we wanted to examine whether MDMA affected ER stress and exodosis 52, 53 . We observed a marked increase in BiP expression in hyperthermic conditions when MDMA was present, although ERdj4 and ASNS levels were not changed by the presence of MDMA (Fig.…”

“…In addition, the present study found that BITC increased the size of the acidic vesicles in AGS cells and upregulated a major lysosomal surface protein; LAMP2, suggesting lysosomal swelling. Enlarged lysosomes and LAMP2 upregulation are known to contribute to lysosomal dysfunction ( Wang et al ., 2018 ; Li et al ., 2020 ).…”
