Macromolecular delivery of 5-aminolaevulinic acid for photodynamic therapy using dendrimer conjugates

Abstract: Intracellular porphyrin generation following administration of 5-aminolaevulinic acid (5-ALA) has been widely used in photodynamic therapy. However, cellular uptake of 5-ALA is limited by its hydrophilicity, and improved means of delivery are therefore being sought. Highly branched polymeric drug carriers known as dendrimers present a promising new approach to drug delivery because they have a well-defined structure capable of incorporating a high drug payload. In this work, a dendrimer conjugate was investiga… Show more

“…The first step towards nano-sized PS formulations were made through the preparation of dendritic [80,81], glyco [82,83], amino acid [84] and peptide [85] functionalized porphyrins. Initially, many of these studies were just aimed at increasing the water solubility, formulation properties and targeting of the PS [86].…”

“…Dendrimers are hyperbranched polymers that have attracted considerable interest in oncology for drug delivery, owing to their ability to carry high drug-payloads and their well-defi ned macromolecular structure [ 11 ] . Recently, a dendrimer containing 18 ALA molecules coupled through biodegradable ester linkages was synthesised and tested in vitro and in vivo [ 12,13 ] . Those studies indicated a more effi cient and sustained PpIX production than ALA at drug equivalent doses after i.p.…”

“…21 5-Aminolevulinic acid (5-ALA)-conjugated dendrimers showed efficient PDT in cell lines. 22 A number of studies have tested the PS-dendrimer complexes; however, very few studies have tested the formulations in in vivo tumor regression studies. 23,24 Commercially available fourth generation poly (amidoamine) (PAMAM) dendrimers are used in this study.…”

“…51 EGFR-specific peptide GE-111 peptide and PS (Pc4) were tested successfully against HER2-specific cells using polymeric nanoparticles and PDT. 52 5-ALA attached to dendrimers by ester bonds was more efficient on A431 cell lines compared to free 5-ALA. 22 Third-generation aryl ether dendritic porphyrins, dendrimers of porphyrin monomers, were found to be very efficient in PDT applications. 53 Several studies used phage display methods to identify efficient tumor-targeting peptides.…”
