Macrophage metabolic regulation in atherosclerotic plaque

Abstract: Metabolism plays a key role in controlling immune cell functions. In this review, we will discuss the diversity of plaque resident myeloid cells and will focus on their metabolic demands that could reflect on their particular intraplaque localization. Defining the metabolic configuration of plaque resident myeloid cells according to their topologic distribution could provide answers to key questions regarding their functions and contribution to disease development.

“…Monocytes and macrophages are regarded as the main actors in atherosclerosis because of their ubiquitous involvement during all stages of atherosclerosis [ 4 , 9 ]. Considerable amounts of evidence support that atherosclerosis is a chronic inflammatory disease starting with the entry of monocytes from peripheral blood into the endothelium: rolling, adhesion, activation and migration.…”

“…After their migration into the intimal space, monocytes are rapidly confronted with various microenvironments, with different available metabolites that will direct their differentiation into M1 or M2 macrophages. Glucose, fatty acid and amino acid metabolism are thought to modulate macrophage phenotype and function at the different stages of atherosclerosis (for review, see [86]). For example, specific glucose fluxes have been shown to be utilized in M1-and M2-type macrophages in glycolysis and the pentose phosphate pathway [87].…”
