Macrophages inhibit and enhance endometriosis depending on their origin

Abstract: Macrophages are intimately involved in the pathophysiology of endometriosis, a chronic inflammatory disorder characterized by the growth of endometrial-like tissue (lesions) outside the uterus. “…It remains unknown what is different about the lesions that progress or are maintained long-term, and future work should draw comparisons between resolving versus non-resolving mice. Recently, we identified a ‘protective’ population of monocyte-derived large peritoneal macrophages (LpM) in our menses mouse model of endometriosis that limit the development of lesions ( Hogg et al, 2021 ). Using gain- and loss-of-function experiments, we demonstrated that depletion of this population increases development of lesions, whereas reprogramming the peritoneal niche such that embryo-derived LpM are ablated and the niche is repopulated afresh with monocyte-derived LpM, leads to decreased lesion development.…”
