Abstract: Malignant hyperthermia (MH) is a pharmacogenetic disorder characterized by acute hypercatabolic reactions in muscles in response to the triggering effects of certain drugs (used mainly during anaesthesia ~ and stresses.

“…8 Later that decade, Beverly Britt, MD, conducted pioneering research on the occurrence and manifestations of MH. 9 This seminal work demonstrated that MH was precipitated by exposure to potent inhalational anesthetics and the paralyzing agent succinylcholine but not to other anesthetics. The MH syndrome was determined to be inherited in an autosomaldominant manner and was independent of ethnicity.…”

“…17 Through the 1970s, MH mortality was close to 80%. 9 When anesthesiologists were made aware of the signs of MH and the steps required to treat it, mortality rates declined. 18 However, a specific antidote, dantrolene sodium, was not approved by the Federal Drug Administration until 1979.…”

“…Although MH syndrome has been associated with congenital myopathies [2,9] such as Duchenne muscular dystrophy, myotonia congenita and central core disease, frequently no signs of myopathy are apparent in patients susceptible to MH [2], In the majority of cases, the mode of inheritance is clearly a single, autosomal dominant gene [5], In other families there appears to be a multigenic, reduced penetrance pattern of inheritance [10]. In addition, the MH trait does not exhibit uniform severity, but rather the MH reaction is a spectrum ranging from mild to severe (variable expressivity).…”

“…This value was defined as the caffeinespecific concentration (CSC) for the strips exposed to caffeine only, and caffeine-specific concentration-halothane (CSC-H) for strips exposed to caffeine plus halothane. In our laboratory, the diagnosis of MH susceptibility is made when the CSC value is less than 4.0 mmol/1 caffeine and the CSC-H value is less than 1.0 mmol/1 caffeine [5]. In this study, MH-susceptible muscle was termed caf feine/halothane contracture test-positive.…”
