Malignant tumor cell lines produce interleukin-1-like factor

Abstract: Sixty-four malignant cell lines were examined for interleukin-1 (IL-1) activities in their conditioned medium using thymocyte and fibroblast proliferation assays. Sixteen cell lines showed high IL-1 activity. Comparison of these activities with human IL-1 showed similarity between some biological properties. However there was no correlation between cell origin and IL-1 activity. These results suggest the possibility that most malignant cells may produce an IL-1-like factor.

“…Ability to manipulate the machinery of cell death is an obvious goal of medical research and effect on regulation of apoptosis might lead to new possibilities for cancer treatment [30]. It is well documented that the secondary metabolites of Sinularia sps have induced apoptotic DNA fragmentation and condensation of chromatin in A431 cells [31]. Keeping in view, investigations were conducted for the first time to study the action mechanism of MDTFC on human leukemia THP-1 cells.…”

“…The A431 human vulval-derived epidermoid carcinoma cell line 7) and Nakata oral squamous cell carcinoma cell line 8) were used in this study. These cell lines were maintained in a humidified atmosphere of 5% CO 2 /95% air at 37 C in an RD medium (1:1 mixture of an RPMI1640 medium and Dulbecco's modified Eagle's medium (DMEM; Sigma Co., St. Louis, MO, USA) supplemented with 5% calf serum (CS; Hyclone Laboratories, Logan, UT, USA).…”

“…To obtain information on the structure-activity relationship, we tested the cytotoxicity of this metabolite in comparison with that of the perhydro derivative, DHA, and other common fatty acids by using two human tumor cell lines, A431 and Nakata cells. 7,8) The results are summarized in Table 3; the IC 50 values for ZL, DHA, ZL-H 12 , linoleic acid, and linolenic acid were determined to be 0.23, 0.21, 7.1, 2.2, and 2.2 M, respectively, against A431 cells, and 0.27, 0.24, 3.5, 1.7, and 2.3 M, respectively, against Nakata cells. The low specific activity of ZL in comparison with that of the crude EtOAc extract would have been due to other cytotoxic metabolites in the EtOAc extract, one of which was confirmed to be the carotenoid, peridinin.…”

“…Tumour cells can also activate directly factor X by procoagulant cysteine proteinase, which has been found in some patients with lung, prostate, colon, breast and kidney cancers as well as in patients with leukaemia [34, 35]. Tumour cells can also activate the clotting system indirectly by stimulating mononuclear cells to synthesize and release various procoagulants [36, 37, 38]. …”
