Mammalian Mip/LIN-9 interacts with either the p107, p130/E2F4 repressor complex or B-Myb in a cell cycle-phase-dependent context distinct from the Drosophila dREAM complex

Abstract: Mammalian Mip/LIN-9 is a cell cycle regulatory protein that is negatively regulated by CDK4/cyclin D. It has been demonstrated that Mip/LIN-9 collaborates with B-Myb during S and G 2 /M in the induction of cyclins A and B, and CDK1. The ortholog of Mip/LIN-9 in Drosophila, Mip130, is part of a large multisubunit protein complex that includes RBF, repressor E2Fs and Myb, in what was termed the dREAM complex. A similar complex, although lacking B-Myb, was also described in Caenorhabditis elegans. Here, we demons… Show more

“…Lin-9 associates with B-Myb but not with the pocket proteins in F9 cells Previous studies have shown that Lin-9 forms complexes with either B-Myb or the p107/p130 pocket proteins at different stages of the cell cycle (Schmit et al, 2007;Pilkinton et al, 2007a). Using immunoprecipitation and western blotting, it was evident that B-Myb was coprecipitated with Lin-9 from F9 cell extracts but p107 was not, despite its expression in these cells (Figure 3a).…”

“…Caenorhabditis elegans contains the DRM complex (Harrison et al, 2006) and three reports have identified human complexes, named LINC (or DREAM), whose composition is regulated at distinct phases of the cell cycle (Litovchick et al, 2007;Schmit et al, 2007;Pilkinton et al, 2007a). LINC contains Lin-9, Lin-37, Lin-54, Lin-52 and RbAp48 (the human homologues of Drosophila Mip130, Mip40, Mip120, dLin-52 and Caf1p55, respectively).…”

“…The DREAM (or LINC) complex is a master regulator of mitotic gene expression during the cell cycle (Schmit et al, 2007, reviewed in Blais and Dynlacht, 2007;Litovchick et al, 2007;Osterloh et al, 2007;Pilkinton et al, 2007a;Knight et al, 2009). Several proteins that are involved in chromosome segregation and cytokinesis, such as Bub1 and survivin, are direct targets of DREAM (Osterloh et al, 2007;Schmit et al, 2007;Pilkinton et al, 2007b;Knight et al, 2009).…”

DREAM consists of a fiveprotein core module and associated proteins (Litovchick et al, 2007;Schmit et al, 2007;Pilkinton et al, 2007a;Knight et al, 2009). In quiescent cells DREAM binds to the E2F4 transcription factor and to p130, which is related to the retinoblastoma tumor suppressor protein.…”
