The aim of the study is to evaluate comparatively the cytotoxicity of diclofenac sodium and calcium hydroxide on L929 fibroblasts. L929 fibroblast cells were cultured and grown on Dulbecco modified Eagle’s medium. Intracanal medicaments tested were Diclofenac sodium, 5.0, 7.5, 10.0 mM/ml) and calcium hydroxide. The human fibroblast cell lines cultured in Dulbecco Modified Eagle’s medium were used as control group. Cytotoxicity was evaluated by 3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) assay. The results showed that there was a significant difference in cell viability as compared with the control group (P<0.05). There was no significant difference in the group treated with diclofenac sodium and calcium hydroxide (1.0 mM/ml). However, diclofenac sodium at concentration more than 5 mM/ml was found to be cytotoxic. The study concludes that diclofenac sodium is cytotoxic at 5 mM/ml and above. Therefore, further studies are recommended to establish the antimicrobial efficacy of the medicament. Within the limitations of the study, Diclofenac sodium at concentration more than 5mM/ml was found to be cytotoxic for the cells. The inhibitory concentration (IC50) of Diclofenac sodium at which the cells were viable was found to be 5.2 mM/ml. Further studies should be done to establish the antimicrobial efficacy of the medicament at these concentrations.

Objectives This study was conducted to investigate the microhardness, surface roughness (Ra), and wear behavior of thin occlusal veneers (TOV) fabricated from different injectable composite materials and compare them to a Computer-Aided Design (CAD)/Computer-Aided Manufacturing (CAM) resin-based material. Materials and Methods A 1-mm occusal veneer preparation was done in a mandibular right second molar typodont tooth. The prepared model was duplicated to fabricate 32 replicas and divided into four groups (n = 8). Standard TOV were fabricated either indirectly from Cerasmart blocks, Cerasmart, GC (CS), or directly from Beautifil Injectable X, Shofu (BF), G-ænial Universal injectable, GC (GU), or SonicFill 2, Kerr (SF) using the injection molding technique. All the specimens were subjected to both thermomechanical cyclic loading (TMC) in a chewing simulator. Wear measurement was conducted by three-dimensional (3D) scanning of the veneered models before and after TMC, and the difference in the volume of the sample was recorded as the volumetric material loss due to wear. Ra before and after TMC and Vickers microhardness (VHN) of the tested materials were measured using standardized samples (n = 8). Representative samples from each group were investigated under a stereomicroscope and a scanning electron microscope. Statistical Analysis One-way analysis of variance (ANOVA) was applied to detect the effect of material on VHN and wear. Two-way ANOVA was utilized to examine the impact of material and TMC on Ra. Multiple comparisons between the groups were conducted using Tukey's post hoc test (α = 0.05). The Pearson's correlation coefficient was used to determine the relationship between hardness and wear and between roughness and wear (α = 0.05). Results CS exhibited the highest mean VHN (p ≤ 0.001), followed by GU and SF which were statistically similar (p = 0.883) but significantly higher than BF (p < 0.001). After TMC, GU revealed the lowest Ra and volumetric wear (VW), followed by CS, BF, and SF (p < 0.5). A highly significant correlation existed between Ra and VW (p = 0.001, R 2 = 0.9803). Conclusion The effect of TMC on the surface properties and wear resistance of the investigated TOV is material-dependent. GU injectable TOV are less influenced by TMC than CS milled TOV. In contrast, BF and SF demonstrated significant VW and Ra which might limit their clinical use as TOV.

Background: Erosive acids might create surface flaws and deteriorate the mechanical properties of CAD-CAM materials. This invitro study aimed to investigate the effect of simulated gastric HCl and extrinsic erosive acids on surface microhardness and fracture toughness of CAD-CAM materials. Methods: 400 bar-shaped specimens (17x4x2mm) were prepared from 4 different CAD -CAM materials (n=100/group); monolithic zirconia (Ceramill Zolid HT+, Amanngirbach, Austria), lithium disilicate ceramic (IPS e.max CAD, Ivoclar Vivadent, Liechtenstein), nanohybrid resin composite (Grandio Blocs, VOCO) and polymer-infiltrated glass network (Vita Enamic, VITA Zahnfabrik). Specimens from each material type were further subdivided into 5 groups (n=20) according to the erosive media applied (simulated gastric HCl, white wine, Coca-Cola, orange juice, and artificial saliva that served as a control). Specimens were immersed for 24 hrs in an incubator at 37 ℃, then ultrasonically cleaned in distilled water and air-dried. Half of the specimens were tested for Vickers microhardness (VH) at parameters of 500 gf for 10 s, while the rest of the specimens underwent 3-point flexure till fracture. Fractured surfaces were examined under a scanning electron microscope (SEM) for fracture toughness (KIC) calculation using the quantitative fractographic analysis method. Data collected were statistically analyzed using two-way analysis of variance (α=0.05) after verification of data normality and homogeneity of variances. Results: Erosive media created surface flaws that lowered the surface microhardness of the material and initiated the fracture pattern under different loads according to material type. The material type was a more predominant factor than erosive media that affected either the microhardness or the fracture toughness of CAD-CAM materials. The highest VH and KIC values were found among Ceramill Zolid HT+ groups followed by IPS e.max CAD and Grandio Blocs regardless of the erosive media employed. Erosive media significantly reduced the VH and KIC in VITA Enamic specimens compared to the rest of the material types. Conclusion: All CAD-CAM materials used, except Vita Enamic, showed high resistance against the erosive acids indicating higher longevity of the material in patients frequently exposed to either extrinsic or intrinsic acid.
