Mangiferin from Pueraria tuberosa reduces inflammation via inactivation of NLRP3 inflammasome

Abstract: Recent reports have demonstrated the role of phyto-constituents in modulating inflammatory responses. Mangiferin isolated from Mangifera indica is known to induce potent anti-oxidative, anti-diabetic and anti-inflammatory activity. However, the molecular mechanism of its anti-inflammatory activity is not properly understood. In this study we have isolated Mangiferin from the tubers of Pueraria tuberosa (PT-Mangiferin) and analysed the mechanism of its potent anti-inflammatory effects in LPS stimulated RAW 264.… Show more

“…There are several known causes of inflammation, and the NF-κB/COX-2/iNOS pathway is a known important mechanism 6,7 . Notably, NF-κB controls various stages of inflammation and immune modulation via the regulation of several molecules, including cytokines (e.g., IL-1β, TNF-α), iNOS, and chemokines 32,33 .…”

“…MF displays anti-diabetic, anti-cancer, and anti-infection effects by regulating inflammation, apoptosis, and cell proliferation as well as mediating immunomodulation 16 19 . In addition, it has been reported to influence the activation or expression of several signal cascades, such as NF-κB, Nrf2/HO-1, mitochondria-dependent pathways, and the NLRP3 inflammasome, and to target multiple cytokines and antioxidant enzymes, such as interleukin (IL)-6, tumor necrosis factor (TNF), superoxide dismutase (SOD), and catalase (CAT) 10 12 , 17 , 19 21 . In terms of the central nervous system, Yang et al .…”

“…Previous investigations elucidated that MF markedly suppressed the productions of IL-6, IL-1b, and TNF-a in LPS-triggered primary hepatocytes, BALB/c mice mammary gland, and peritoneal macrophages (Jeong et al, 2014;Pan et al, 2016;Qu et al, 2017a). Furthermore, an earlier research revealed that PT-MF remarkably decreased the expression of Cox-2, iNOS and TNF-a, and the generation of ROS in LPS-evoked RAW 264.7 cells (Bulugonda et al, 2017). In this investigation, we firstly explored the influences of MF treatment alone on the ATDC5 cells.…”
