Mapping and DNA sequence characterisation of the Rysto locus conferring extreme virus resistance to potato cultivar ‘White Lady’

Abstract: Virus resistance genes carried by wild plant species are valuable resources for plant breeding. The Ry sto gene, conferring a broad spectrum of durable resistance, originated from Solanum stoloniferum and was introgressed into several commercial potato cultivars, including 'White Lady', by classical breeding. Ry sto was mapped to chromosome XII in potato, and markers used for marker-assisted selection in breeding programmes were identified. Nevertheless, there was no information on the identity of the Ry sto g… Show more

“…The variant Rysto-like26 found in cultivars Alicja, Bzura and White Lady, as well as the variant Rysto-like16 found in the breeding line PW363, encode a protein with 100% identity to the Ry sto reference protein. One of the two additional variants found in cultivar Alicja, Rysto-like30, showed 100% nucleotide identity to the TMV2 gene in cultivar White Lady, identi ed by ne-scale genetic mapping and characterized by Sanger sequencing in a previous study [25]. TMV2 displayed 95% identity to the reference Ry sto gene published by Grech-Baran et al 2020 [19].…”

“…The possible reason might be different plant material used in this study (the tetraploid potato cultivar Alicja) and the previous study (the dihaploid clone Alicja) [19] or just sequencing errors, as such regions are prone to errors. Different Rysto-like variants identi ed in different cultivars tested, as well as additional variants identi ed in cultivar Alicja in this study, may re ect a different origin of the gene (Figure S2) [15,19,25]. The source of the Ry sto reference gene is the dihaploid clone Alicja, which was obtained from the tetraploid potato cultivar Alicja via parthenogenesis [19,26].…”

“…The variant Rysto-like26 found in cultivars Alicja, Bzura and White Lady, as well as the variant Rysto-like16 found in the breeding line PW363, encode a protein identical to the Ry sto reference protein. One of the two additional variants found in cultivar Alicja, Rysto-like30 , showed 100% nucleotide identity to the TMV2 gene in cultivar White Lady, identified by fine-scale genetic mapping and characterized by Sanger sequencing in a previous study [ 27 ]. TMV2 displayed 95% identity to the reference Ry sto gene published by Grech-Baran et al 2020 [ 19 ].…”

“…Payette Russet has ‘extreme resistance’ governed by the NLR gene, Ry sto , which has been introgressed from the wild potato species, S. stoloniferum , into commercial cultivars [ 43 ]. The Ry sto locus has been mapped to the distal arm of potato chromosome 12, which contains four clusters of resistance genes [ 44 , 45 ]. An NLR protein encoded by the Ry sto gene recognizes and binds to the coat protein encoded by strains of PVY and PVA [ 43 ].…”
