Cyperus has been commonly used as a multi-use medicinal plant in folk medicine worldwide. The objectives of our study were to determine the different metabolites in the Cyperus conglomeratus Rottb. methanol extract, and to assess its in vivo gastroprotective effect in ethanol-induced gastric ulcer model in rats. Serum levels of galactin-3 and TNF-α were employed as biochemical markers. To pinpoint for active agents, comprehensive metabolites profiling of extract via UPLC-qTOF-MS/MS was employed. A total of 77 chromatographic peaks were detected, of which 70 were annotated. The detected metabolites were categorized into phenolic acids and their derivatives, flavonoids, stilbenes, aurones, quinones, terpenes, and steroids. Rats were divided into six groups; healthy control, ulcer control, standard drug group, and 25, 50, 100 mg/kg of C. conglomeratus treated rats. Pre-treatment with C. conglomeratus alcohol extract significantly reduced galactin-3, and TNF-α in ethanol-induced ulcer model at 25, 50, and 100 mg/kg. Further histopathological and histochemical studies revealed moderate erosion of superficial epithelium, few infiltrated inflammatory cells, and depletion of gastric tissue glycoprotein in the ulcer group. Treatment with the extract protected the gastric epithelial cells in a dose-dependent manner. It could be concluded that C. conglomeratus extract provides significant gastroprotective activity in ethanol-induced gastric ulcer and ought to be included in nutraceuticals in the future for ulcer treatment.

Anti‐inflammatory and antioxidant effects of Apium graveolens L. extracts mitigate against fatal acetaminophen‐induced acute liver toxicity

In the present work, antioxidant activity, total phenolics (TP), and total flavonoids (TF) contents of aqueous and methanol extracts of celery were determined, in addition to untargeted metabolites profiling its methanol celery root extract (MCRE) via UPLC-MS. Although MCRE exhibited the lowest TPC and TFC levels, it presented the most potential hydroxyl radical quenching effect using electron paramagnetic resonance spin trapping technique. Treatment of Acetaminophen-induced hepatotoxicity (AAH) rats with MCRE lowered serum levels of AST, ALT, ALP, TNF-α, and IL-1β significantly. Additionally, MCRE significantly increased total antioxidant capacity (TAC) and glutathione (GSH) levels relative to AAH rats. Strikingly, Kaplan-Meier survival analysis of all groups revealed a 100% prevention of acetaminophen-induced mortality of rats by MCRE pretreatment (100 mg/kg/day). MCRE prevented AAHassociated severe weight loss and elicited normal behavior in the rescued rats. Our results suggest that pretreatment with MCRE can mitigate against overdosed acetaminophen-induced acute liver failure and warrant further investigations on the potential of postinjury intervention.

BackgroundAlzheimer's disease (AD) is a prevalent neurodegenerative disorder without an effective cure. Natural products, while showing promise as potential therapeutics for AD, remain underexplored.AimsThis study was conducted with the goal of identifying potential anti‐AD candidates from natural sources using Caenorhabditis elegans (C. elegans) AD‐like models and exploring their mechanisms of action.Materials & MethodsOur laboratory's in‐house herbal extract library was utilized to screen for potential anti‐AD candidates using the C. elegans AD‐like model CL4176. The neuroprotective effects of the candidates were evaluated in multiple C. elegans AD‐like models, specifically targeting Aβ‐ and Tau‐induced pathology. In vitro validation was conducted using PC‐12 cells. To investigate the role of autophagy in mediating the anti‐AD effects of the candidates, RNAi bacteria and autophagy inhibitors were employed.ResultsThe ethanol extract of air‐dried fruits of Luffa cylindrica (LCE), a medicine‐food homology species, was found to inhibit Aβ‐ and Tau‐induced pathology (paralysis, ROS production, neurotoxicity, and Aβ and pTau deposition) in C. elegans AD‐like models. LCE was non‐toxic and enhanced C. elegans' health. It was shown that LCE activates autophagy and its anti‐AD efficacy is weakened with the RNAi knockdown of autophagy‐related genes. Additionally, LCE induced mTOR‐mediated autophagy, reduced the expression of AD‐associated proteins, and decreased cell death in PC‐12 cells, which was reversed by autophagy inhibitors (bafilomycin A1 and 3‐methyladenine).DiscussionLCE, identified from our natural product library, emerged as a valuable autophagy enhancer that effectively protects against neurodegeneration in multiple AD‐like models. RNAi knockdown of autophagy‐related genes and cotreatment with autophagy inhibitors weakened its anti‐AD efficacy, implying a critical role of autophagy in mediating the neuroprotective effects of LCE.ConclusionOur findings highlight the potential of LCE as a functional food or drug for targeting AD pathology and promoting human health.
