Mapping of calpastatin and three microsatellites to porcine chromosome 2q2.1-q2.4

Abstract: Three polymorphisms were identified in a 1.6-kb fragment of the porcine calpastatin (CAST) gene and these polymorphisms were used for genetic linkage mapping. Linkage analysis revealed significant linkage of CAST to five microsatellites previously mapped to porcine chromosome 2; these microsatellites were S0010, S0226, SWI4, Sw395 and Sw776. A somatic cell hybrid panel was used to determine the chromosomal localization of CAST and the microsatellites S0091, S0226 and Sw395. All of these were localized to the r… Show more

“…The primer sequence and PCR conditions to amplify the product of the CAST gene (GenBank Accession No. EU137105.1) were applied following the method detailed by Ernst et al (1998). For genotyping, genetic variants were identified by the PCR-RFLP method using two restriction Ngu et al 13783 enzymes namely HinfI and MspI (MBI, Fermentas).…”

“…Total DNA was isolated from 50 mg of pig tissue using a commercially available High Pure PCR Template Kit (Roche Diagnostics GmbH, Mannheim, Germany), according to the protocol provided by manufacturer. The genotypes at different CAST loci were identified with HinfI, MspI and RsaI restriction enzymes according to the method described by Ernst et al (1998). PCR was performed in a reaction mixture of 20 mL containing 50 ng genomic DNA, standard PCR buffer, 1.5 mM MgCl2, 200 mM of each dNTP, 5 pmol of each primer and 1 U Taq DNA polymerase (Fermentas, Vilnius, Lithuania).…”

“…Unlike calpains, which are coded by at least 14 genes (Kemp et al 2010), calpastatin is coded by only one gene -CAST, mapped on SSC2 in Region 2q2.1-q2.4 (Ernst et al 1998). Ciobanu et al (2004) sequenced complete CAST gene and identified two single nucleotide polymorphisms (SNPs) associated with meat tenderness, namely Arg249 Lys and Ser638Arg.…”

“…Therefore, this gene affects the proteolytic system of meat during the post mort period (KRISTENSEN et al, 2002;ROPKA-MOLIK et al, 2014). The calpastatin gene (CAST) is located on chromosome 2 (2q2.1-2q2.4) (ERNST et al, 1998;DAVOLI et al, 2017). CIOBANU et al (2004) reported an association analysis of a missense mutation (Ser638Arg) in the CAST gene where quantitative trait loci (QTL) for meat quality such as shear force and tenderness, were described.…”
