Mapping of ribosomal DNA and (TTAGGG)n telomeric sequence by FISH in the bivalve Patinopecten yessoensis (Jay, 1857)

Abstract: The chromosome set of Patinopecten yessoensis (Jay, 1857) was characterized using Giemsa staining, DAPI staining and fluorescence in situ hybridization (FISH) with three repetitive DNA probes [18S-28S rDNA, 5S rDNA and telomeric (TTAGGG) n ]. DAPI staining showed that AT-rich regions were located on the centromere of almost all chromosomes and interstitial banding was not observed. FISH showed that 18S-28S rDNA spread over the short arms of two subtelocentric chromosome pairs and 5S rDNA was located on the lon… Show more

“…The situation in families of the other subclass of Bivalvia (Pteromorphia) is somewhat more variable. Major rDNAs occur in only one chromosome pair in most Ostreidae species analyzed (Cross et al 2003; and in half of the species of Pectinidae Insua et al 2006;Odierna et al 2006), but not in Hinnites distortus (Lopez-Piñon et al 2005), Patinopecten yessoensis (Huang et al 2007b), Argopecten irradians Huang et al 2007a), and A. purpuratus (Gajardo et al 2002). However, in Mytilidae, major rDNA was found in one (Brachidontes pharaonis, Vitturi et al 2000;B.…”

“…Most species of Pectinidae also show a single cluster of 5S rDNA López-Piñón et al 2005;Insua et al 2006;Huang et al 2007b), but Aequipecten opercularis shows two clusters. For Argopecten irradians one (Huang et al 2007a) and two clusters have been described. The presence of two clusters of 5S rDNA is also the case in all species of Ostreidae (Cross et al 2005;Wang et al 2005).…”

“…Labeled probes were purified by SanPrep PCR products purify kit (Sangon Biotech) and then resolved at a concentration of 5-10 ng/µl in a hybridization solution of 2×SSC (sodium chloride/sodium citrate), 50% deinoized formamide and 10% dextran sulphate. 18S-28S rDNA probe were obtained by PCR amplification and labeled with biotin-16-dUTP based on previous study (Huang et al 2007b). …”

“…Cytogenetic studies of the Yesso scallop showed it possessed 38 chromosomes with a karyotype formula of 3m+5sm+8st+3t (Huang et al 2007b). Yet, most of these studies were mainly focusing on the multi-copy genes or sequences such as rDNA sequences and histones genes (Huang et al 2007b, Zhang et al 2007).…”
