Mapping of the secretome of primary isolates of mammalian cells, stem cells and derived cell lines

Abstract: Within a mammalian organism, the interaction among cells both at short and long distances is mediated by soluble factors released by cells into the extracellular environment. The secreted proteins may involve extracellular matrix proteins, proteinases, growth factors, protein hormones, immunoregulatory cytokines, chemokines or other bioactive molecules that have a direct impact on target cell phenotype. Stem cells of mesenchymal, adipose, neural and embryonic origin, fibroblast feeder cells as well as primary … Show more

“…In recent years it has been increasingly accepted that their regenerative effects are mainly mediated by their secretome [4][5][6]. The secretome, which comprises the proteins released by cells, tissues or organisms, has been shown to be crucial to the regulation of different cell processes [7].…”

“…MSCs have been found in a variety of adult tissues of mesodermal origin, such as bone marrow, adipose tissue, placenta, umbilical cord, dental pulp or synovium [20][21][22][23][24][25] . Although sharing the main characteristics, differences between MSCs from different sources can be found, for instance at the RNA and protein expression profiles levels [26][27][28] . MSCs are considered a promising tool for cell therapy, in particular for inflammatory diseases, based on their immunomodulatory properties and paracrine effects through trophic factors with anti-fibrotic, anti-apoptotic or pro-angiogenic properties [29,30] .…”

“…It also enables determination and discovery of new roads of tissue cross talk and interaction. QMSP has been applied to study secretomes of a variety of cell types and tissues including adipose cells and tissues, mouse embryonic fibroblasts, astrocytes, mesenchymal stem cells, neuronal progenitor cells, kidney, and endothelial cells (Skalnikova et al, 2011). Although, there have been several proteomics reports describing the secretory function of cells from mesenchymal origin, the role of the muscle secretome has remained elusive.…”
