Marine Natural Products, Multitarget Therapy and Repurposed Agents in Alzheimer’s Disease

Abstract: Alzheimer’s disease (AD) is a multifactorial disease characterized by the presence of amyloid plaques, neurofibrillary tangles, and nerve cell death that affects, mainly, older people. After decades of investigation, the search for an efficacious treatment for AD remains and several strategies can be and are being employed in this journey. In this review, four of the most promising strategies, alongside with its most promising agents under investigation or development are highlighted. Marine natural products (… Show more

“…These compounds range from small peptides and enzymes to highly complex secondary metabolites that show bioactivities in physiological systems such as those related to movement, circulation, and respiration. The importance of marine drugs against neurodegeneration is widely recognized [ 32 , 33 ]. In fact, some natural compounds are already in preclinical studies for AD treatment [ 34 ].…”

“…Marine natural products, developed under adverse conditions and containing unusual structures, have been a rich source for the treatment of numerous diseases including AD. A number of marine natural products, together with their derived synthetic analogs, showed good efficacy against AD [ 288 ]. Thus, marine natural products are actively sought after as neuroprotective agents for AD.…”

“…Most investigations have been focused on their effect on MMPs during inflammatory diseases and/or cancer. Due to the relevance of MMPs (especially MMP-2 and MMP-9) in neurodegenerative diseases, marine products could also be useful for the inhibition of MMPs in AD [ 74 ].…”
