Context Swertia mussotii Franch. (Gentianaceae) is a source of the traditional Tibetan medicine, ZangYinChen, and is used to treat chronic hepatitis and many types of jaundice. Objective This study explored the therapeutic effects and mechanism of S. mussotii on non-alcoholic fatty liver disease in diet-induced hypercholesterolaemia. Materials and methods After a week of adaptive feeding, 32 Sprague-Dawley rats were divided into four groups: (1) Control, (2) Control-S, (3) Model, and (4) Model-S. During the 12 experimental weeks, we established the Model using a high-fat diet. Control-S and Model-S were given 1.0 g/kg S. mussotii water extract via gavage starting in the fifth week until the end of experiment. Results When compared with Model rats, the S. mussotii water extract led to a reduction in high-density lipoproteins (43.9%) and albumin (13.9%) and a decrease in total cholesterol (54.0%), triglyceride (45.6%), low-density lipoproteins (8.6%), aspartate aminotransferase (11.0%), alanine aminotransferase (15.5%), alkaline phosphatase (19.1%), total protein (6.4%), and glucose (20.8%) in serum. A reduction in three cytokines (IL-1β, IL-6, and TNFα) was detected. Histopathological examination showed that liver steatosis was significantly relieved in S. mussotii -treated high-fat diet rats. S. mussotii also caused a downregulation in the expression of TLR4 (43.2%), MyD88 (33.3%), and a decrease in phosphorylation of NF-κB. Discussion and conclusions Our findings indicate that S. mussotii may act as a potential anti-inflammation drug via inhibition of the TLR4/MyD88/NF-κB pathway. Further in vivo and in vitro studies are needed to validate its potential in clinical medicine.

Porifera are essential components of marine ecosystems, providing valuable ecological functions. Traditional approaches to estimating sponge growth and biomass are destructive and often not suitable for certain morphologies. The implementation of new innovative techniques and nondestructive methodologies have allowed for a more sustainable approach. In this study, a population of Sarcotragus foetidus Schmidt, 1982 (Demospongiae, Dictyoceratida, Irciinidae), thriving inside the Portofino Marine Protected Area, was monitored using Structure from Motion photogrammetry over a period of 6 years, from September 2017 to October 2023. Of the 20 initial individuals, only 12 were still in place during the last monitoring, indicating 40% mortality. Through photogrammetry, the overall volume change and biomass production were estimated to be 9.24 ± 5.47% year−1 and 29.52 ± 27.93 g DW year−1, respectively, indicating a general decreasing trend between 2021 and 2023. Signs of necrosis were observed in some individuals, potentially related to the high temperature occurring during summer 2022 and 2023. Considering the current climate crisis, long-term monitoring efforts must be made to better understand the dynamics of this species, and photogrammetry has the potential to be a versatile monitoring tool that will contribute to the standardization of methodologies for sponge growth studies.

Sponges and their associated microorganisms have tremendous potential in the medical world to be explored. However, improper exploration will cause habitat destruction and cost much. The application of metabolomics to sponges and their associted microbes can be the best solution in their exploration, which involves a combination of chemical profiling and multivariate analysis (chemometrics). In this , we 47 genera of sponges and 24 genera of their associated microorganisms that were studied with a metabolomic approach until July 2021. The sponges most often studied in metabolomics-related research are Geodia, Xestospongia, Agelas, and Aplysina. There are four analytical techniques that are often used, namely, liquid chromatography-mass spectrometry, polymerase chain reaction, nuclear magnetic resonance, and gas chromatography-mass spectrometry, in determining the chemical/genomic profile. There are eight chemometric analyses that are often used in metabolomic applications on sponges and their associated microbes, namely, similarity analysis, principal component analysis, hierarchical cluster analysis, partial least square, partial least square-discriminant analysis, orthogonal projections to latent structures, orthogonal projections to latent structures-discriminant analysis, and linear discriminant analysis. The most widely used metabolomic applications for sponges and their associated microbes for the last decade are for identification and dereplication purposes, for quality control purposes, and for the purpose of linking chemical profiles and bioactivity patterns. The purpose of other metabolomic applications, namely, to determine bioavailability to quantitatively determine bioactivity, and to test safety and toxicity, has yet to be carried out because research on sponges and their associated microbes is still around the discovery of new compounds and quality control.
