mascRNA and its parent lncRNA MALAT1 promote proliferation and metastasis of hepatocellular carcinoma cells by activating ERK/MAPK signaling pathway

Abstract: MALAT1-associated small cytoplasmic RNA (mascRNA) is a cytoplasmic tRNA-like small RNA derived from nucleus-located long noncoding RNA (lncRNA) metastasis-associated lung adenocarcinoma transcript 1 (MALAT1). While MALAT1 was extensively studied and was found to function in multiple cellular processes, including tumorigenesis and tumor progression, the role of mascRNA was largely unknown. Here we show that mascRNA is upregulated in multiple cancer cell lines and hepatocellular carcinoma (HCC) clinical samples.… Show more

“…Our previous studies showed that non-coding RNAs, including miRNAs and lncRNAs (long non-coding RNAs), play essential roles in skeletal muscle development ( Xie et al, 2018 ; Liu et al, 2021 ; Tan et al, 2021 ). LncRNAs are a class of non-coding RNAs greater than 200 nucleotides in length with limited or no protein-coding capacity that possess complex spatial structures and diverse functions ( Chen and Carmichael, 2010 ; Xie et al, 2021a ). Numerous studies have shown that lncRNAs play a significant role in biological functions, such as epigenetic modification, mRNA transcription, splicing, stability and translation ( Lan et al, 2021 ).…”

“…In addition, the MALAT1 expression level is negatively correlated with the survival rate in cancer patients 28 . MALAT1 induces cell proliferation and metastasis via the MAPK/ERK and PI3K/AKT signaling pathways in retinoblastoma and ovarian cancer, respectively 29 , 30 , and it is known to enable the high expression of the key oncogene MYC in thymic epithelial tumors 31 . Interestingly, in HepG2 cells, MALAT1 was also found in mitochondria, and its knockdown limited ATP synthesis and tumor cell invasion 32 .…”

“…Although previous literature has verified that the ERK phosphorylation can promote apoptosis and reduced the doxorubicin's efficacy in osteosarcoma, [26][27][28] effect of MALAT-1 on the ERK/ MAPK pathway is not consistent. For example, some literature indicated that MALAT-1 activates ERK/MAPK pathway in hepatocellular carcinoma 29 and cardiomyocytes, 30 while another literature showed that MALAT-1 inhibits ERK/MAPK pathway in the nervous system. 31 Indeed, the potential mechanisms resulting in drug resistance are complex.…”
