-Penaeid acute viremia (PAV) or white spot disease (WSD) causes mass mortality in cultured penaeid shrimp. Despite many histopathological studies, the principal lesion in the diseased shrimp remained unclear. Here we reevaluated the histopathology and ultrastructure of moribund kuruma prawn Penaeus japonicus artificially infected with penaeid rod-shaped DNA virus (PRDV) as a pathogen of PAV. We showed that PRDV infected cardiac satellite cells, primary myocardial cells and myocardial cells of the heart, where the nuclei of these cells displayed virus propagation that resulted in severe myocardial necrosis. Myocardial necrosis was suggested to be a primary cause of the viremia of PAV.

Although the Janus kinase-signal transducer and activator of transcription (JAK-STAT) signaling pathway is part of the antiviral response in arthropods such as Drosophila, here we show that white spot syndrome virus (WSSV) uses a shrimp STAT as a transcription factor to enhance viral gene expression in host cells. In a series of deletion and mutation assays using the WSSV immediate-early gene ie1 promoter, which is active in shrimp cells and also in insect Sf9 cells, an element containing a STAT binding motif was shown to be important for the overall level of WSSV ie1 promoter activity. In the Sf9 insect cell line, a specific protein-DNA complex was detected by using electrophoresis mobility shift assays (EMSA) with the 32 P-labeled STAT binding motif of the WSSV ie1 promoter as the probe. When recombinant Penaeus monodon STAT (rPmSTAT) was overexpressed in Sf9 cells, EMSA with specific antibodies confirmed that the STAT was responsible for the formation of the specific protein-DNA complex. Another EMSA showed that in WSSV-infected P. monodon, levels of activated PmSTAT were higher than in WSSV-free P. monodon. A transactivation assay of the WSSV ie1 promoter demonstrated that increasing the level of rPmSTAT led to dose-dependent increases in ie1 promoter activity. These results show that STAT directly transactivates WSSV ie1 gene expression and contributes to its high promoter activity. We conclude that WSSV successfully annexes a putative shrimp defense mechanism, which it uses to enhance the expression of viral immediate-early genes.

Very high mortalities of juvenile Metapenaeus ensis occurred in the seedling production at public sea-farming centers in west Japan during 1994 and 1995. Moribund shrimp exhibited no apparent external signs except small obscure white spots on the body, and were proved to be free from any causative bacteria, fungi and parasites. Infection trials reproduced the disease in both healthy juvenile M. ensis and Penaeus japonicus. The disease was characterized by nuclear hypertrophy of the necrotic cells in various organs and tissues originated from meso-and ectoderm, such as cuticular epidermis and hematopoietic tissue. Enveloped ovoid-shaped virus particles were observed in such hypertrophied nuclei in sectioned preparations and in the hemolymph in negatively stained preparations. Penaeid rod-shaped DNA virus (PRDV)specific DNA fragments were detected by PCR method. These results indicate that the mass mortalities were caused by penaeid acute viremia (PAV).
