Mass spectrometry as a diagnostic and a cancer biomarker discovery tool: Opportunities and potential limitations

Abstract: Serum proteomic profiling, by using surfaced-enhanced laser desorption/ionization-time-of-flight mass spectrometry, is one of the most promising new approaches for cancer diagnostics. Exceptional sensitivities and specificities have been reported for some cancer types such as prostate, ovarian, breast, and bladder cancers. These sensitivities/specificities are far superior to those obtained by using classical cancer biomarkers. In this review, I concentrate more on questions that cast doubt on the results repo… Show more

“…Albumin acts as a sink or reservoir for these molecules and greatly prolongs their half-life from minutes to days (25). Hence, newer assays that rely on the detection of multiple (uncharacterized) peptide peaks are being developed to probe this peptide space (26); however, it is not clear whether these current peptide fingerprint assays are sufficiently reproducible and robust for clinical use (27)(28)(29).…”

“…22,23 Approximately a quarter of cellular proteins secreted or shed within the CNS are present in the intracellular milieu. 24 Therefore, it is reasoned that the CSF proteome can provide valuable information about the CNS in disease. [25][26][27][28][29] Recent advances in proteomics combine multidimension analyses of the global protein composition with mass spectrometric identification.…”
