Matrix metalloprotease-2 and -9 concentration and activity in serum and culture medium samples: a methodological reappraisal

Abstract: Our data indicate that different values for MMP concentrations can be obtained using different analytical methods, even if they are intrinsically reliable. This suggests that methodological differences should be taken into account when comparing MMP results from different studies.

“…Even though MMP-2 is weakly inducible, it is aberrantly up-regulated in the tumor microenvironment and serum concentration is, then, dramatically high (Colotti et al, 2007): this last aspect has been addressed for the MMP-2 screening as a relevant diagnostic and prognostic marker (Patel et al, 2011). Epidemiologic studies suggest MMP-2 quantification as prognostic marker in patients with colorectal cancer, ovarian cancer and thyroid cancer (Kwon et al, 2011;Rajoria et al, 2011).…”

“…Because serum concentration of MMP-2 was significantly elevated in tumor patients, MMP-2 was suggested as a diagnostic and prognostic marker [156, 157]. On the other hand elevated MMP-9 levels in the serum seem to be even more relevant values, because in healthy individuals under physiological conditions MMP-9 is hardly detectable [158].…”

“…Moreover, MMPs bound to their tissue inhibitors (TIMPs) and the MMPs/TIMPs complexes can be found in high concentrations in peripheral circulation . These complexes are recognized to different extents by different immunometric and enzymatic assays for MMPs ; thus, different analytical methods could lead to different results and these methodological differences have to be taken carefully into account when clinical/experimental data are compared. While immunometric systems recognize pro‐ and active MMPs together, the enzymatic assays, exploiting the biological properties of the active form of MMPs, are able to independently evaluate both the pro‐ and the active form of MMPs, after a preanalytical treatment of a biological sample .…”

“…The aim of this study was to perform a reappraisal of quantitative gel zymography technique for gelatinases, considering MMP‐9 as a representative example. A comparison was made between the results obtained with zymography and those obtained by a widely used ELISA, whose analytical features had been previously evaluated by us .…”
