ABSTRACT. We carried out a case-control study to evaluate the role of MMP-9 -1562 C/T (rs3918242) genetic polymorphism in the risk of pre-eclampsia in Chinese pregnant women. Between March 2013 and January 2015, 107 pregnant women with pre-eclampsia were recruited from the Second Affiliated Hospital of Zhengzhou University. Genotyping of MMP-9 was carried out using polymerase chain reactionrestriction fragment length polymorphism assay. Unconditional logistic regression analyses revealed that women with the CT genotype (OR = 1.81, 95%CI = 1.04-3.11) exhibited significantly higher risk of preeclampsia than those with the wild-type CC genotype. Moreover, individuals carrying the T allele were at higher risk of pre-eclampsia than those carrying the C allele; the adjusted OR (95%CI) was 1.62 2 C. Sun et al. Genetics and Molecular Research 15 (3): gmr.15038355 (1.06-2.47). In conclusion, our study demonstrated that the MMP-9 -1562 C/T polymorphism plays an important role in the development of pre-eclampsia in Chinese women. Further studies with large sample sizes are greatly needed to confirm our findings.

Abnormal matrix metalloproteinase (MMP)-9 levels may have a role in hypertensive disorders of pregnancy. We examined whether MMP-9 genetic polymorphisms (g.À1562C4T and g.À90(CA) 13À25 ) modify plasma MMP-9 and tissue inhibitor of metalloproteinase (TIMP)-1 levels and the responses to antihypertensive therapy in 214 patients with preeclampsia (PE), 185 patients with gestational hypertension (GH) and a control group of 214 healthy pregnant (HP). Alleles for the g.À90(CA) 13À25 polymorphism were grouped L (low) (o21 CA repeats) or H (high) (X21 CA repeats). Plasma MMP-9 and TIMP-1 concentrations were measured by enzyme-linked immunosorbent assay. Plasma MMP-9 concentrations were not affected by genotypes or haplotypes in HP and PE groups, except for the g.À90(CA) 13À25 polymorphism: GH patients with the LH genotype for this polymorphism have higher MMP-9 levels than those with other genotypes. The T allele for the g.À1562C4T polymorphism and the H4 haplotype (combining T and H alleles) are associated with GH and lack of responsiveness to antihypertensive therapy in GH. The H2 haplotype (combining C and H alleles) was associated with lack of responsiveness to antihypertensive therapy in PE, but not in GH. In conclusion, our results show that MMP-9 genetic variants are associated with GH and suggest that MMP-9 haplotypes affect the responsiveness to antihypertensive therapy in hypertensive disorders of pregnancy.

Abstract. Phagocytosis of malaria pigment (hemozoin) induces increased activity of matrix metalloproteinase (MMP)-9, an endopeptidase involved in cytokine regulation. In this study, we examined whether a common functional MMP-9 promoter polymorphism (rs3918242) affects Plasmodium falciparum infection in pregnancy. Eighteen percent of Ghanaian primiparae carried the minor T allele. It was associated with reduced odds of placental hemozoin and of placental as well as peripheral blood parasitemia. The results indicate that a common MMP-9 polymorphism protects against placental malaria indicating that this endopeptidase is involved in susceptibility to P. falciparum.Pregnant women are a particular risk group for infection with Plasmodium falciparum and malaria. Although commonly asymptomatic at high endemicity, malaria in pregnancy may cause anemia, abortion, stillbirth, low birth weight (LBW), and preterm delivery (PTD), and contributes to high infant mortality. The increased susceptibility of pregnant women, particularly primigravidae, is largely due to parasites expressing specific variants of the P. falciparum erythrocyte membrane protein-1. Parasite adhesion via these variant surface proteins results in the sequestration of infected red blood cells (RBCs) in the placental intervillous space. Sequestration frequently is accompanied by local hemozoin (malaria pigment) deposition and accumulation of inflammatory cells, including monocytes/macrophages. Specific immune mechanisms targeting the pregnancy-associated parasites, particularly parasite-specific antibodies, are low in primigravidae. Only with successive pregnancies, these are acquired, and infection risk and manifestation decrease.
