Matrix Metalloproteinase (MMP) 9 Transcription in Mouse Brain Induced by Fear Learning

Abstract: Background: Matrix metalloproteinase 9 is involved in fear-associated memory formation wherein transcriptional regulation is poorly known. Results: Overexpression and promoter binding activity of AP-1 factors regulate MMP-9 transcription, preceding elevated enzymatic activity in mouse brain. Conclusion: c-Fos and c-Jun AP-1 components positively regulate MMP-9 transcription in fear learning. Significance: The novel tools and approaches in vivo allowed us to explore MMP-9 transcription in mouse brain.

“…4A). NFκB is a known regulator of mmp9 expression in other systems (Eberhardt et al, 2002;Ganguly et al, 2013). Using a previously published NFκB reporter line, Tg(NFκB:gfp), we observed an increase in NFκB activity after tail transection by 6 hpw ( Fig.…”

“…MMP-9 basal activity was not affected in either of the GSK-3β-modified mice, possibly because of the low levels of constitutive MMP-9 or because its activity is affected in specific subfields of the hippocampus [41, 42, 64, 65]. …”

“…Meanwhile, MMP‐9 gene expression is activated through a pathway that involves extracellular signal‐regulated kinases to activate serum response factor to stimulate the expression of the c‐ fos gene and then trigger MMP‐9 gene expression with the aid of AP‐1 transcription factor (Ganguly et al . ; Kuzniewska et al . ).…”
