Measurement of Allergens Associated with Dust Mite Allergy

Abstract: Sensitive radioimmunoassays were developed to measure the two groups of major allergens (Der I and Der II) of Dermatophagoides pteronyssinus and Dermatophagoides farinae, in which radiolabeled protein A served as a general tracer. Glass rods covalently coupled with F(ab′)2 fragments of rabbit antiserum IgG were incubated first with mite or house dust extracts, and then with affinity-purified rabbit antibodies. The bound allergen-antibody complex was detected with a 125I-labeled protein A.… Show more

“…House dust mites of the Dermatophagoides species ( Dermatophagoides pteronyssinus and Dermatophagoides farinae ) are the most important causative factor associated with various allergic diseases among all the indoor allergens [1–3]. Group 1 (Der p 1 and Der f 1) and group 2 (Der p 2 and Der f 2) allergens are considered the major house dust mite allergens [4–7]. IgE‐binding activity for both group 1 and group 2 allergens is detected in the majority of sera from patients allergic to house dust mites.…”

“…The major causative factors associated with various allergic diseases are house dust mite ( Dermatophagoides farinae and Dermatophagoides pteronyssinus ) allergens [16–19]. The major allergens from house dust mite are classified as group 1 (Der f 1 and Der p 1, M r 25 000) or group 2 (Der f 2 and Der p 2, M r 14 000) [20–25]. Der f 2 is the major group 2 allergen from D. farinae [26,27], and consists of 129 amino acid residues with marked homology to Der p 2 [28], the group 2 allergen from D. pteronyssinus .…”

“…Recent data provide evidence that the allergens amplify allergen-induced cytokine expression (5). Indeed, several studies demonstrated that Der p 1 stimulates release and expression of cytokines in human airway epithelial cells (6), including IL-13 in cord blood mononuclear cells (7) and IL-2 and IL-4 in a cord blood T-cell phenotype with atopic dermatitis (8). Although the release of cytokines by allergen stimulation has been studied, details of the intracellular signaling mechanism, including the secretion of cytokines, remain unclear.…”
