Measurement of gastric emptying during and between meal intake in free-feeding Lewis rats

Abstract: A new scintigraphic measurement technique is described that allows accurate assessment of gastric emptying in between as well as during a number of successive meals. Measurements were made every minute of food intake, gastric nutrient filling, and gastric emptying over a 6 h, 40 min period in conscious, free-feeding, loosely restrained rats. Before receiving access to the food, the animals had been deprived for a period of 31 h. Over the full duration of the experiment, an average rate of gastric emptying of 2… Show more

“…On days 3, 5, 7, and 9, we detected 0, 46.52, 178.59, and 486.21 g of AL/g in the stom-ach contents at 8 h after the last dose. The increasing amount of drug observed remaining in the stomach over the course of dosing shows that gastric emptying is inhibited (Kaplan et al, 1992;Li et al, 1998b;van der Velde et al, 1999). The inhibition of gastric emptying progresses from mild on day 5 to severe inhibition on day 9.…”

“…The rates of gastric emptying within a meal (i.e., when gastric contents accumulate as a result of ongoing ingestion) have been previously shown to greatly exceed the rate of nutrient emptying after meal termination (15,48). Thus, in the hyperphagic OLETF rat, the greater magnitude of gastric fill due to increased meal size may result in increased emptying relative to LETO controls.…”

“…Figures 1-2 show how cells located in the intestinal epithelium sense and respond to nutrients. Under normal conditions, roughly a third of all calories consumed exit the stomach prior to the end of the meal[9-11]. What remains is gradually metered out over time via negative feedback from the intestine.…”
