Measurement of teicoplanin by liquid chromatography-tandem mass spectrometry: development of a novel method

Abstract: Background: Teicoplanin is an antibiotic used for the treatment of endocarditis, osteomyelitis, septic arthritis and methicillinresistant Staphylococcus aureus. Teicoplanin is emerging as a suitable alternative antibiotic to vancomycin, where their trough serum levels are monitored by immunoassay routinely. This is the first report detailing the development of a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for measuring teicoplanin in patients' serum. Methods: An Acquity TM UPLC (ultra-pres… Show more

“…The comparability between this immunoassay, the QMS ® teicoplanin, and our newly developed LC-MS method is moderate. However, the correlation coefficient r = 0.856 (r 2 = 0.73) observed by us resembles the previously reported correlation between teicoplanin measurements by another LC-MS method and FPIA (r 2 = 0.86) [19]. Possible reasons for the relatively poor correlations include different cross-reactivities of the antibodies with the different teicoplanins as well as interferences with the immunoassay.…”

“…To the best of our knowledge, there are currently only two other published applications of LC-MS to TDM of teicoplanin [19,20]. Our method is the first method using online extraction based on turbulent flow chromatography.…”

“…The existing available methods for assaying TCP are high-performance liquid chromatography (HPLC), microbiological assay and liquid chromatography-mass spectrometry. [6][7][8] Among them, HPLC is the main measurement method for TCP because of its high separation e±ciency and good selectivity. However, these methods involve a tedious variety of reagents or lengthy run times, which are unsuitable for rapid measurement of TCP in the production process or in blood.…”

“…Mass transitions determined to be suitable for the quantitation of teicoplanin (940.6 m/z → 316.5 m/z, and 940.6 m/z → 298.3 m/z) were consistent with the literature. 13 Erythromycin was used as an internal standard (734.5 m/z → 158.5 m/z, and 734.5 m/z → 576.9 m/z). Analytes were separated on an Alltech™ Alltima™ Reversed-Phase HPLC Column, 20 mm × 2.1 mm, with 3 μm particles.…”
