Measurements of amino acid transport in internally dialyzed giant axons

Abstract: It is shown that the axoplasmic composition of acidic and neutral amino acids can be controlled effectively by the method of internal dialysis. Direct assay for specific binding and measurement of diffusion coefficients in axoplasm show that there is no significant binding or compartmentalization of amino acids. The dependence of amino acid efflux on substrate concentration can be measured under well-defined, true steady-state conditions. The taurine efflux-concentration relation in the Myxicola giant axon con… Show more

“…It is not known what role glycine and glutathione have in the cephalopod olfactory system. Glycine acts at several synapses of the squid central nervous system (Caldwell and Lea, 1978; Horn, 1986; Vinogradova et al, 2002; Lima et al, 2003), but, because there is no clear role for glycine in our model organisms, we cannot draw any conclusions about the role of glycine in squid. There were two classes of cells in squid OE that had high glutathione labeling (Classes 5 and 6), and were morphologically identified as a subset of type 2 cells.…”

“…Recently, Horn (1986) has developed a modification using a much more permeable dialysis tubing. For the present study, we have used a dialysis tubing with a nominal molecular-weight cut-off of 6,000 (Spectrapor HF MWCO 6000, Spectrum, Los Angeles, CA).…”
