Abstract:Increasing work on mitochondria is focused on the dynamic aspects of this organelle, a key part of which is regulation of the mitochondrial genome (mtDNA). Here we describe a protocol that selectively incorporates bromodeoxyuridine into mtDNA for the measurement of mtDNA turnover, synthesis or supercoiling via an adapted immunoblot and Southern blot protocol.
