Mechanical loading stimulates bone angiogenesis through enhancing type H vessel formation and downregulating exosomal miR‐214‐3p from bone marrow‐derived mesenchymal stem cells

Abstract: Exosomes are important transporters of miRNAs, which play varying roles in the healing of the bone fracture. Angiogenesis is one of such critical events in bone healing, and we previously reported the stimulatory effect of mechanical loading in vessel remodeling. Focusing on type H vessels and exosomal miR-214-3p, this study examined the mechanism of loading-driven angiogenesis. MiRNA sequencing and qRT-PCR revealed that miR-214-3p was increased in the exosomes of the bone-losing ovariectomized (OVX) mice, whi… Show more

“…PlGF has a variety of functions, including promoting osteogenesis and angiogenesis, and plays an essential role in the regulation of osteogenic-angiogenic interactions by mechanical stimulation (McCoy et al, 2013). It has also been found that mechanical stimulation can stimulate H vessel formation and VEGF secretion by downregulating exosomal miR-214-3p from MSCs (Wang et al, 2021).…”

“…Mechanical loading can increase YAP expression in vascular endothelial cells and enhance skeletal blood perfusion, 61 resulting in increased angiogenic activity 62 and type H vessel (CD31 hi EMCN hi ) formation near the femoral growth plate. 63 Type H vessels are spatially coupled with osteogenic cells. In bone defect repair, type H vessels were observed within the defect and surrounded by a large number of skeletal stem cells.…”

“…Bone repair and remodeling is a complicated process which involves angiogenic and osteogenic interaction [44] . Herein, our study showed that hUC-MSCs-sEVs could be uptaken by BMSCs and then facilitated the proliferation, migration of BMSCs, indicating that BMSCs migrated to the defect area and further proliferated which induced by hUC-MSCs-sEVs.…”
