Mechanical reduction of the intracanal Enterococcus faecalis population by Hyflex CM, K3XF, ProTaper Next, and two manual instrument systems: an in vitro comparative study

Abstract: Canal instrumentation by all the file systems significantly reduced the intracanal Enterococcus faecalis counts. ProTaper Next was found to be most effective in reducing the number of bacteria than other rotary or hand instruments.

“…Both ProTaper Next and Twisted File promoted a higher bacterial reduction than the ProTaper Universal and manual technique. The result found for the ProTaper Next is in accordance with a study by Tewari et al (11) and Krokidis at al. (20), who also observed a better performance of this system compared to other automated systems and manual technique.…”

“…The plate culture method was used in this study because of its simplicity and efficiency (5,7,(8)(9)(10)(11)(13)(14)(15)(16)(17)(18)(19). Molecular methods has been highlighted because of their high sensibility to detect microorganisms, but such a feature would not be necessary in this study as there was no total absence of bacterial growth in the collection.…”

“…The importance to evaluate the tip size of this instrument is based on the current tendency towards single-file systems. Premolars have been used in several endodontics studies (5,6,14), but it is necessary instruments with diameters larger than tip 25. Moreover, molar is a tooth which requires the greatest amount of endodontic treatment (15).…”

“…In this study each root canal was inoculated with 0.5 ml of E. faecalis, with density adjusted to the turbidity of 0.5 McFarland standard (1.5 x 10 8 bacteria/ml) according to Blanscet et al, (17) , this was followed by incubation period 48 hrs.to allow deeper penetration of E. Faecalis into dentinal tubules according to Tewari et al (18) .…”
