Genital tracts of male Lewis rats were transected at various levels to determine whether this may influence antisperm antibody response. Adult male rats underwent bilateral transection of the vas deferens (group I, n -9), mid-epididymis (group 11, n -lo), and efferent duct (group III, n -9).Group IV (n = 10) underwent a sham operation. Sera were collected by retro-orbital puncture before the operative procedure and monthly for 3 months postprocedure. Sperm-reactive immunoglobulins IgG, IgA, and IgM were measured individually as well as combined in serum by enzyme-linked immunosorbent assay (ELISA) using lithium diiodosalicylate (LIS)-solubilized washed rat caudal epididymal sperm. The maximal immune response was seen in all groups at 2 months postprocedure. Antibody response defined as the net ELISA absorbance reading for the combined immunoglobulin group were (mean f SEM): group I = 120 * 16, group I1 -156 f 23, group 111 -190 f 20, and group IV -116 * 22. The highest antibody response was noted in the efferent duct group, which was statistically (p < .05) greater than the sham-operated and vas deferens groups. In the efferent duct group the highest immunoglobulin response was observed in the IgG class, which was significantly higher (p < .05) than the IgA and IgM classes. The transection of the male genital tract at different levels leads to variation in antisperm antibody response and that sperm located at different sites along the genital tract may differ in their autoantigenic potential.

In this study, we aimed to examine the effect of L-arginine, tadalafil, and their combination for prevention of the ischemia reperfusion injury after testis torsion in rats. Methods: A total of 40 adult, male Sprague-Dawley rats were allocated into five groups. Three hours of left testicular torsion was performed in each group, excluding the control group. While the ischemia reperfusion (I/R) group had no treatment, I/R + Arg group received L-arginine, I/R + Td group received tadalafil and I/R + Arg + Td group received tadalafil and L-arginine 30 minutes before the detorsion. Then the left testis was untwisted for four hours of reperfusion. After bilateral orchiectomy, lipid peroxidation (LPx) and glutathione (GSH) activities were examined in testicular tissue. Spermatogenesis was evaluated with Johnsen's score. Results: LPx levels of the I/R group were found to be significantly higher than for groups that received drugs for both testes (p<0.001). GSH levels of the combination group were higher than I/R group in ipsilateral testis (p<0.01) and it was significantly higher than other groups for contralateral testis (p<0.001 for I/R group, p<0.01 for I/R + Arg, p<0.05 for I/R + Td). Mean Johnsen's score of the I/R group was found to be significantly lower than treatment groups in ipsilateral testis (p<0.001 for I/R + Arg + Td group, p<0.01 for other treatment goups) and contralateral testis (p<0.001). The mean Johnsen score of the combination group was significantly higher than that of other treatment groups in ipsilateral testis (p<0.05) and it was significantly higher than in the I/R + Td group in the contralateral testis (p<0.05). Conclusions: L-arginine, tadalafil, and combination of these two molecules showed protective effect against ischemia/reperfusion injury for both testes after unilateral testis torsion.

The effect of unilateral testicular torsion on the contralateral testis and the fertility rate was studied in Charles River adult rats. Animals were divided into groups that underwent a sham operation or torsion and ligation of the left testicular vessels followed by orchiectomy after 24 h, orchiectomy after 48 h, release of the ligature after 24 h, release of the ligature after 48 h, and no further treatment following ligation. Another group of animals underwent unilateral orchiectomy. After 8 weeks animals were allowed to mate and were sacrificed 2 weeks later. The results did not point to either histological alterations in the contralateral testis or impairment of fertility in any group of treatment compared with the control.
