Mechanism of Enhanced Nifedipine Dissolution by Polymer-Blended Solid Dispersion through Molecular-Level Characterization

Abstract: We investigated the effect of polymer composition on nifedipine (NIF) dissolution through molecular-level characterization of NIF/hypromellose (HPMC)/Eudragit S (EUD-S) ternary solid dispersions. The dissolution rates and molecular states of NIF and polymers were evaluated in NIF/HPMC/EUD-S spray-dried samples (SPDs) with different polymer compositions. Blending of HPMC and EUD-S improved the dissolution property of each polymer. Moreover, polymer blending enhanced NIF dissolution from the NIF/polymer SPD with… Show more

“…Nifedipine (NDP) is a typical poorly water-soluble drug that is pharmaceutically used as a calcium channel blocker for the treatment of cardiovascular diseases [ 1 ]. Because of its poor water solubility (<5 μg/mL) [ 2 ], its dissolution and subsequent oral absorption are limited [ 3 , 4 ]. Therefore, its solubility must be enhanced via various formulations [ 2 , 3 , 4 , 5 ].…”

“…Because of its poor water solubility (<5 μg/mL) [ 2 ], its dissolution and subsequent oral absorption are limited [ 3 , 4 ]. Therefore, its solubility must be enhanced via various formulations [ 2 , 3 , 4 , 5 ]. The term “dispersion” refers to a drug in a polymer matrix or inert carrier [ 6 ] as a solid dispersion caused by the use of a meltable compound followed by solvent evaporation or melting solvent methods.…”

“…In the absence of API recrystallization in the dissolution media, the degree of supersaturation appears to be dependent on the degree of vinyl pyrrolidone substitution. This dependency on vinyl pyrrolidone content for ketoprofen dissolution is an interesting observation which may be explained by the concept of congruent dissolution of drug and polymer, which is known to be aided by the presence of intermolecular interactions [40,41]. This argument is strengthened by the evidence of hydrogen bonding between PVP and ketoprofen which has been provided by the FTIR data.…”

“…Therefore, a membrane permeability study in the well-established epithelial cellular models will be investigated in our further research. Furthermore, utilizing fast-dissolving excipients as carriers is an effective way to enhance drug dissolution [45]. The solubility of LM in water was significantly higher than that of MCC, which might result in higher bioavailability in vivo.…”
