Mechanism of validamycin A inhibiting DON biosynthesis and synergizing with DMI fungicides against Fusarium graminearum

Abstract: Deoxynivalenol (DON) is a vital virulence factor of Fusarium graminearum, which causes Fusarium head blight (FHB). We recently found that validamycin A (VMA), an aminoglycoside antibiotic, can be used to control FHB and inhibit DON contamination, but its molecular mechanism is still unclear. In this study, we found that both neutral and acid trehalase (FgNTH and FgATH) are the targets of VMA in F. graminearum, and the deficiency of FgNTH and FgATH reduces the sensitivity to VMA by 2.12‐ and 1.79‐fold, respecti… Show more

“…Beneficial microorganisms include bacteria, actinobacteria, fungi, and viruses, which are closely associated with plant health and safety (Munir et al, 2021 ; Ahmed et al, 2022 ). Microbial metabolites, such as validamycin and streptomycin, are the most widely used biological pesticides at present (Copping and Menn, 2000 ; Bian et al, 2021 ). Trichoderma was first isolated from soil in 1794 and it has been a well-known biocontrol resource since then.…”

“…Streptomycin is a broad-spectrum drug effective against both Gram-negative aerobic bacteria and staphylococci. Another aminoglycoside compound is validamycin [ 52 ], which has been used to control the rice sheath blight caused by Rhizoctonia solani for over 50 years in China, with no validamycin-resistant isolates reported in the field [ 52 , 53 ].…”

“…In the current study, FGSG_01959 encoding a cytochrome P450 protein associated with farnesyl pyrophosphate in F. graminearum, was signi cantly up-regulated with phenamacril treatment. Farnesyl pyrophosphate (FPP) converted from acetyl-CoA from the isoprenoid biosynthetic pathway, is the immediate precursor of DON biosynthesis [53]. FgCYP51B (FGSG_01000), encoding cytochrome P450-51 protein, and FGSG_09786 encoding NADPH-cytochrome P450 reductase, were downregulated with carbendazim treatment.…”
