Mechanisms of Fibroblast Activation in the Remodeling Myocardium

Abstract: Purpose of Review Activated fibroblasts are critically implicated in repair and remodeling of the injured heart. This manuscript discusses recent progress in the cell biology of fibroblasts in the infarcted and remodeling myocardium, highlighting advances in understanding the origin, function and mechanisms of activation of these cells. Recent Findings Following myocardial injury, fibroblasts undergo activation and myofibroblast transdifferentiation. Recently published studies have suggested that most activa… Show more

“…9 Wnt pathway has revealed new points of intervention that may lead to novel drug targets for small molecular weight compounds. Wnt signal is relatively silent in adult heart tissue, 7 but re-activated after a variety of cardiac injuries ranging from acute ischaemic insult to chronic pressure overloading.…”

“…8 Abnormal Wnt pathway activation contributes to transforming growth factor-beta (TGF-β)-induced fibrotic remodelling in numerous adult fibrotic diseases including cardiac fibrosis after MI. 9 Wnt pathway has revealed new points of intervention that may lead to novel drug targets for small molecular weight compounds. 10,11 However, how Wnt signalling is dysregulated in post-ischaemic heart needs further investigations.…”

“…According to our results, expression of Cthrc1 peaks at 14dpi and decreases thereafter. Besides the potential for defining the population of IRCF, this dynamic suggest that expression of Cthrc1 may be responsible for promoting the fibrotic response after MI, an essential part of the healing process 3,34 . Targeted ablation of other markers of activated CF such as Postn 9,35 or Fstl1 28 have been associated with increased mortality after MI.…”

“…Cardiac fibroblasts (CF) represent only 10% of the total number of cells in the myocardium; however, they play a critical role in the structural and mechanical maintenance of the heart 1,2 . After myocardial injury, provoked by acute ischemia, CF become activated orchestrating a fibrotic response that leads to generation of a collagen scar, which prevents cardiac rupture 3,4 . The absence of distinct markers for the identification of CF has traditionally hampered our understanding of the fibroblast roles in cardiac homeostasis.…”
