MEF2 transcription factors promotes EMT and invasiveness of hepatocellular carcinoma through TGF-β1 autoregulation circuitry

Abstract: Invasion and metastasis is the main causes leading to the death of hepatocellular carcinoma (HCC) patients. However, the underlying mechanism is still to be explored. Transforming growth factor β1 (TGF-β1) is a stronger inducer of HCC cell invasion. However, the downstream effector of TGF-β1 that promotes HCC invasion is still unknown. In this study, we found that PI3K/Akt activation takes place following the stimulation of TGF-β1. The inhibition of PI3K/Akt activation abolished epithelial-mesenchymal transiti… Show more

“…More interestingly, Pokemon and miR-125 form a molecular circuit that can contribute to the development of HCC. In fact, a similar regulatory auto-circuit has been identified in HCC cells, such as TGF-β/MEF2D [17]. Our finding reinforces that notion that malignant tumors can utilize auto-regulatory circuit to support its initiation and progression.…”

“…These results indicated that there is an auto-regulatory circuit consisting of Pokemon and MEF2D, which can promote the invasion of HCC cells. Such loop-like networks were also observed to exist in HCC [9] and in some other types of human cancers, including glioma [12].…”

“…Natural compounds can be used for the suppression of these two genes. A reference reported that oleanolic acid can suppress the invasion of HCC by downregulating MEF2D expression [9]. Other strategies are needed to be developed to silence the expression of these two HCC-related oncogenes.…”

“…MEF2C and MEF2D have been well established to play important roles in the progression of hepatocellular carcinoma (HCC) by promoting VEGF signaling pathway and increasing cell proliferation rates, respectively [1,2]. The invasion and metastasis of HCC were also demonstrated to be enhanced by the MEF2 protein overexpression via promoting a TGF-β1 autoregulation circuitry [3]. In addition to solid tumors, leukemia was also verified to be associated with MEF2 family of transcription factors [4][5][6].…”
