Several characteristics from sparkleberry (V. arboreum) would be useful if transferred to highbush blueberry cultivars, but to date all direct crosses have failed. The purpose of this research was to see whether crosses of diploid V. darrowii and diploid V. fuscatum (both section Cyanococcus) with diploid V. arboreum (section Batodendron) could be made and to assess the possibility of transferring traits from V. arboreum into cultivated blueberries of section Cyanococcus. Many crosses were successful and easy to make. Crosses were more productive when V. arboreum was the pollen parent, but some hybrids were also obtained with V. arboreum as seed parent. Pollen stainability of F 1 (V. darrowii 9 V. arboreum) hybrids was low, ranging from 0.9 to 23.7%. Numerous meiotic abnormalities were found in the F 1 hybrids. These led to a high percentage of incomplete and unstained tetrads and probably to the production of unreduced gametes.

Diploid Vaccinium darrowii Camp has been used in breeding tetraploid southern highbush blueberry (Vaccinium corymbosum L.) as a source of reduced chilling requirement, adaptation to hot, wet summers, and resistance to leaf diseases. V. darrowii in Florida is quite variable, but most crosses have involved only one V. darrowii clone, Fla. 4B. The use in breeding of a wider range of V. darrowii accessions would provide beneficial diversity in the blueberry cultivated gene pool. The purpose of this research was to determine the functional 2n gamete frequency of numerous V. darrowii genotypes when crossed with tetraploid V. corymbosum, and to study the pollen fertility and backcross ability of the interspecific (V. darrowii × V. corymbosum) hybrids to V. corymbosum. Crosses between diploid V. darrowii and tetraploid highbush blueberry cultivars had low fruit set compared with the V. darrowii × V. darrowii and highbush × highbush crosses. The unusually high number of hybrids per pollinated flower (HPF) in certain 4x-2x or 2x-4x crosses was attributed to high functional 2n gamete production in certain V. darrowii genotypes. Diploid Vaccinium fuscatum Aiton and diploid V. darrowii × V. fuscatum hybrids, when crossed with southern highbush blueberry cultivars, were equally productive of hybrids whether used as male or female parents. Variation in frequency of functional 2n gametes in V. darrowii, expressed as high HPF, was present within plants (megaspores vs. microspores) and among V. darrowii plants. Of the 114 interspecific (V. darrowii × V. corymbosum) hybrids studied, 106 had pollen stainability >50%. This indicated that most of these hybrids were tetraploid, because triploid blueberries, like most triploid plants, are highly sterile. Twenty-two V. darrowii × V. corymbosum hybrids were backcrossed to tetraploid highbush blueberry cultivars. Fruit set was variable, but large populations of vigorous hybrids were obtained. Lower fruit set was associated with hybrids that had lower pollen fertility. It should be possible to obtain plants of cultivar quality in a few generations of backcrosses.

One synthetic hexaploid clone (Hex-DT) was produced by placing 3-node stem cuttings of triploid blueberry clone FL 81-19 [V. corymbosum (4x) x V. elliottii (2x)] on blueberry micropropagation medium supplemented with 0.6% agar, 24.6 μM 2ip, and 0.02% colchicine. The most effective chromosome-doubling was achieved with colchicine over 6 days, for 6 h per day. Pollen stainability and pollen germination in the hexaploid Hex-DT were 42.2% and 13.0%, respectively, versus 87.9% and 50.9% in Hex-F1, a hexaploid F1 hybrid between V. ashei (6x) x FL 81-19 (3x). The number of seedlings produced per flower pollinated with V. ashei pollen in Hex-F1 was similar to the number obtained from V. ashei x V. ashei crosses, and at least twice the number produced by Hex-DT pollinated with V. ashei pollen. Observed mean numbers of univalents, bivalents, trivalents, quadrivalents, and hexavalents per PMC at metaphase I in Hex-DT were 1.29, 20.30, 2.31, 2.63, and 2.11, respectively. No univalents were observed in Hex-F 1, but the mean frequencies of bivalents, trivalents, quadrivalents, and hexavalents were 29.14, 0.87, 1.91, and 0.58, respectively. Irregularities such as chromosome lagging and unequal disjunction were observed at both anaphase I and II in Hex-DT. Anaphases I and II in Hex-F1 were normal with few irregularities. The high fertility observed in Hex-F1 suggests a high level of homology among the three species contributing to its makeup.
