Melatonin Administration Accelerates Puberty Onset in Mice by Promoting FSH Synthesis

Abstract: Although melatonin has been extensively studied in animal reproduction, the mechanism of melatonin in puberty remains elusive. This study was designed to explore the effect of intraperitoneal administration of melatonin on puberty onset in female mice. The injection of melatonin into postnatal days 10 mice at a dose of 15 mg/kg accelerated the puberty onset in mice. Mechanistically, there was no difference in physical growth and serum Leptin levels after melatonin administration. Meanwhile, the serum levels of… Show more

“…High oxygen consumption makes it better adapted to the cold field environment and matches its nocturnal characteristics ( 72 ). Melatonin is a neuroendocrine hormone that affects the growth, and reproduction of mammals ( 73 , 74 ). During the night (especially between 11:00 p.m. and 5:00 a.m.), blood melatonin levels increase 3-10 times and the secretion of melatonin peaks ( 75 ).…”

“…And the mRNA levels of GnRH and GnRHr, key genes in GnRH signaling in the hypothalamus that control FSH synthesis, were unchanged. It can be hypothesized that the positive effect of melatonin on FSH synthesis is not mediated by the hypothalamus, but is directly accelerated by acting on the pituitary gland to accelerate the onset of puberty [ 28 ]. Melatonin can cross the blood–brain barrier, so we choose to prolong melatonin receptor sensitivity by slowly absorbing it through subcutaneous injection.…”

“…By breaking down endocrine barriers, melatonin can accelerate the start of puberty and improve ewes’ reproductive capabilities [ 27 ]. Young female mice were given a substantial dose of melatonin (15 mg/kg) intraperitoneally, which expedited the start of puberty [ 28 ]. However, melatonin implantation before puberty in ewes delayed the onset of puberty by 4 weeks [ 29 ].…”

“…The existing body of research suggests that during the chronotype and the circadian timekeeping system change during the puberty, including endogenous rhythm period and sensitivity to environmental time cues ( 49 ). The results of several studies have established that melatonin promotes follicle-stimulating hormone in the pituitary gland and increases serum estrogen levels, thereby accelerating the onset of puberty ( 50 ). Those studies indirectly support a potential role of TIPIN in AAM.…”
