Melatonin prevents acute kidney injury in severely burned rats via the activation of SIRT1

Abstract: Acute kidney injury (AKI) is a common complication after severe burns. Melatonin has been reported to protect against multiple organ injuries by increasing the expression of SIRT1, a silent information regulator that regulates stress responses, inflammation, cellular senescence and apoptosis. This study aimed to investigate the protective effects of melatonin on renal tissues of burned rats and the role of SIRT1 involving the effects. Rat severely burned model was established, with or without the administratio… Show more

“…Protective actions of melatonin in microglial cell lines stimulated by LPS were reduced by EX527 . Renal tissue of severely burned rats was protected by melatonin in multiple ways, including antioxidant and anti‐inflammatory actions, including downregulation of TNFα and IL‐1β, effects that were blocked by EX527 . However, in the same study, downregulation of the anti‐inflammatory IL‐10 was also described as well as a counteracting effect by the sirtuin blocker, findings that would require further elucidation.…”

“…Recently, SIRT1 was demonstrated to inhibit inflammatory reactions 6. Additionally, we previously showed that SIRT1 regulates the effect of melatonin on kidney protection against inflammation reactions in severely burned rats [19]. Previously study demonstrated that SIRT1 is a direct target of miR-138, participating in cancer and axon regeneration [16,20,21], but whether miR-138 participates in inflammation by targeting SIRT1 is still unclear.…”

“…Previous findings have reported a rise in the expression of SIRT1 in cells and animal models after melatonin treatment . The SIRT1 inhibitor was used to verify that SIRT1 plays a pivotal role in several functions of melatonin including protecting against kidney injury and myocardial ischemia‐reperfusion injury . However, it is still largely unknown whether melatonin‐induced up‐regulation of SIRT1 is responsible for its inhibitory effect on mitochondrial fission.…”
