“…Domains critical for PLC activity are found in urchin PLC: sea urchin PLC-δ contains Ser522, Lys438, Lys440, and Arg549 that are important for human PLC activity [19,33]. Based on structural/functional studies of human and sea urchin PI-3K, domains involved in catalysis (phox homology (PX) and C2 domains) are quite similar and likely mediate similar protein-protein interactions and/or binding to membrane lipids [22,73]. All targeted enzymes thus contain critical domains that have been conserved throughout subsequent evolution, implicating similar catalytic functions in echinoderms and vertebrates.…”

“…Thus, many BAR domains selectively associate with membranes of high curvature due to the shape of their concave surface and the greater ability for facilitating electrostatic interactions with the highly curved membranes. (18), and C2 domains (13). Recently, it was also demonstrated that lipid-binding domains could be regulated by pH.…”

“…The specificity in C2 domain targeting arises in the surface loops, which are variable in amino acid sequence and conformation and most often involved in lipid binding. Also of functional consequence is a cationic patch in the concave face of the bsandwich, termed the cationic b-groove, which varies in size and electrostatics among C2 domains (13). Cationic b-grooves have been shown to bind ceramide-1-phosphate (36) as well as PIs including PI(4,5)P 2 (37).…”

“…They consist of 130 residues and share a common fold composed of two four-stranded β-sheets arranged in a compact β-sandwich connected by surface loops and helices (1)(2)(3)(4). Many of these C2 domains have been demonstrated to function in a Ca 2+ -dependent membrane-binding manner and hence act as cellular Ca 2+ sensors.…”

“…Here, the use of a freemounting system has enabled us to capture an intermediate state of Ca 2+ binding to the C2A domain of rabphilin 3A that suggests a different mechanism of ion interaction. We have also determined the structure of this domain in complex with PI(4,5)P 2 and IP 3 at resolutions of 1.75 and 1.9 Å, respectively, unveiling that the polybasic cluster formed by strands β3-β4 is involved in the interaction with the phosphoinositides. A comparative study demonstrates that the C2A domain is highly specific for PI(4,5)P 2 /PI (3,4,5)P 3 , whereas the C2B domain cannot discriminate among any of the diphosphorylated forms.…”
