Mepanipyrim haptens and antibodies with nanomolar affinity

Abstract: Mepanipyrim is an anilinopyrimidine fungicide used worldwide for crop protection. With the aim of developing useful immunoreagents for mepanipyrim immunoanalysis, two new functionalized derivatives were prepared and antibodies were generated. Affinity and specificity were assessed by direct and indirect competitive ELISA using homologous and heterologous conjugates. Although all antibodies were selective for the target analyte, the immunizing hapten structure was revealed as a determinant for high-affinity ant… Show more

“…[23][24][25] In summary, the antibodies obtained with hapten FXn showed superior affinity and specificity to fluxapyroxad than those derived from hapten FXb, thus evidencing the relevance of the linker position in the binding properties of the generated antibodies, as also demonstrated for other low molecular weight compounds. [26][27][28] For further cELISA development, two combinations of immunoreagents were selected according to the lowest IC 50 value and moderate slope of the inhibition curve, one for the direct format and the other for the indirect format. The optimum concentration of each immunoreagent was determined in order to obtain A max values between 1 and 2 absorbance units.…”

“…Some ELISAs were developed for the detection of mepanipyrim and its related anilinopyrimidine fungicides. [16][17][18][19] They were sufficiently sensitive for mepanipyrim detection but their reactivity with the propanol type was not considered. This study re-ports that monoclonal antibodies (MoAbs) reacted with mepanipyrim and/or its propanol type were prepared by improving the hapten design and that the direct competitive (dc)-ELISAs based on their MoAbs could be applied to residue analysis in vegetables.…”

“…[12,13] QDs have many excellent properties including narrow and symmetric emission spectrum, large Stokes shifts, and low background noise, and the spectral overlap is reduced which are advantageous for detection of fluorescent signals. [18][19][20][21][22] MIP based on QDs as an optosensing material have attracted much attention in recent years, and many methods for selective and sensitive detection of norepinephrine, [23] tetrabromobisphenol A, [24] tocopherol [25] and other analytes [26] have been reported. [17] However, most of the physiological environment is hydrophilic, hydrophobic QDs are difficult to use.…”

“…Based on these properties, QDs have been utilized as fluorescence probes to detect molecules, ions, cells, enzymes, and proteins. Methamidophos (MAP) is a major OPs that has been widely used in fruits and vegetables.…”
