“…PLFA C16:1ω5 was approx. 2.4‰ more 13 C-depleted than roots (Walder et al 2013 ; Ven et al 2020 ) and NLFA C16:1ω5 was approx. 3.7‰ more 13 C-depleted than roots (Ven et al 2020 ).…”

In contrast, AM hyphae have previously been shown to be continuously 13 C-enriched by around 5‰ (Walder et al 2012 , and this study) relative to plant leaves, a pattern also found for other mycorrhizas (ECM) (e.g.…”

“…The neutral lipid fatty acid (NLFA) 16:1ω5 was used as the AM fungal biomass indicator. It has been shown to be more sensitive as an AM fungal signature and correlate better than PLFA 16:1ω5 with other indicators of AM fungal biomass, such as hyphal length density and root colonisation (Olsson et al 1995;Olsson et al 1998;Ven et al 2020).…”

“…Fourteen seedlings of Zea mays L. (a C 4 -plant, variety 'Tom Thumb') per mesocosm were planted on the 21st of June 2017 after their roots were submerged in spore-based inoculum of the AMF Rhizophagus irregularis (Symplanta®). Mesh bags, often used to study AMF (see, e.g., Ven et al, 2020) because they exclude ingrowth of roots but are permeable for fungal hyphae (30 μm mesh, size 13 cm × 3 cm × 2 cm, average soil density 1.497 g cm −3 ), filled with C free white river sand (heated for 5 hr at 550 C to remove any remains of organic matter) were buried vertically into the top soil of each mesocosm 1 week before planting. One mesh bag per mesocosms was harvested at the end of the experiment, which was 10 weeks after planting.…”

“…Mycorrhizal root colonization and hyphal length followed the procedure described by Ven et al (2020). Mycorrhizal root colonization was verified in August by sampling roots from four plants per mesocosm.…”
